The NCOA1 primers were designed from well conserved-sequence region of homologous genes in human and mouse , a fragment was amplified from swine NCOA1 gene. PCR-RFLP procotol was employed to type 454 sows from 3 different foreign breeds in ZhengHong, the effects of them on sow litter size were analyzed. And the fragment was sequenced and analyzed. The main results are as follows:1. Through PCR-RFLP genotype detection, there are three genotypes in Landrace and Large White: AA, AB and BB, the frequency of AA genotype are far high than AB and BB genotypes; but there was only AA genotype found in Duroc, AB and BB genotypes were not detected. And in the whole sow population, the frequency of AA genotype was 0.7455,far high than the others. and the frequency of A allele (0.8613) are far more higher than B allele(0.1387).2. The correlation between NCOA1 gene and litter size were analyzed by using SAS software: the effects of NCOA1 polymorphism on swine litter size are significant, and there is a AA>AB>BB trend in TNB and NBA, the difference between genotypes are significant, the TNB and NBA of AA genotypes are the highest in Landrace and Large White , BB genotypes are the lowest. The TNB and NBA of AA genotype are 2.66 and 1.57 higher than BB genotype in Large White respectively, 2.28 and 2.07 higher in Landrace respectively. It shows that NCOA1 gene have great effects on litter size, A allele is an advantageous gene.3. The sow parity have great effect on litter size, the TNB and NBA of first parity are lower than still parity, and the average litter size in still parity are significantly higher than first parity (P<0.01); the differences between genotypes in still parity are significant, The TNB and NBA of AA genotype are significantly higher than BB genotype(P<0.01). in first parity, the differences between genotypes are not significant in Landrace (P>0.05 ) , but significant in Large White(P<0.05).
