Pig reproductive traits is a type of important economic traits, reproductive performance directly affect the economic benefit of the pig industry. Research has shown that estrogen receptor (ESR) gene and follicle-stimulating hormone beta (FSHβ) genes are the major genes that mainly effect porcine reproductive performance. In this study polymerase chain reaction (PCR) - RFLP method had been used to detect the genetic variation of the two genes and effect to the litter size of the four Anhui local pig breeds, discussed the application strategies of both gene to the protection and utilization of the Anhui local pig breeds.In this study, Polymerase chain reaction (PCR) and PCR- RFLP technology had been used to research the ESR, FSH beta gene polymorphisms in the four local pig breeds of Anhui province (Anqing liubai pig, Dingyuanhei pig, Wannanhei pig and Wannanhua pig). Results show that the ESR genes in Anqing liubai pig and Dingyuanhei pig had been detected in all three genetic type (AA, BB and AB), however in Wannanhei pig and Wannanhua pig only two genotypes( AA and AB) , no BB genotype; FSH beta genes in Wannanhei pig and Wannanhua pig had been detected in in all three genetic types(CC, CD and DD), and in Anqing liubai pig and Dingyuanhei pig only two genotypes( CC and CD) were detected, no DD genotype. By theχ2 compatibility test, the ESR loci in Anqing liubai pig are in Hardy-Weinberg balance, but the FSH beta loci are not; while the result in Dingyuanhei pig, Wannanhei pig and Wannanhua pig is on the contrary.In the four tested populations, the correlation between FSH beta,ESR genotype effect and reproductive trait are not consistent tendency that reported, and some even contrary and achieve significant level. In Anqing liubai pig tested, all the ESR genotypes effect were not significant (P>0.05), but exist the tendency of AA>BB>AB, while in Wannanhua pig exist widely AA>AB, and the both genotype effect in sows differ significantly (P<0.05), that shows A alleles is the dominant genes in both pig breeds. But in Dingyuanhei pig, the ESR genotypes effect exist the tendency of BB >AB >AA, the total number born and number born alive of sows in BB type are significantly differ AB and AA type(P<0.05),while in Wannanhei pig, there are no significant difference between the AA and AB genotype effect to litter size traits(P>0.05),but follow the trend of AA < AB, that shows the B alleles is the dominant genes in both pig breeds.In the research of FSH beta gene loci found that: in the Anqing liubai pig tested group, there is no significant difference(P>0.05) between genotype CC,CD and the four traits assoiated with the number of born, while the genotype effect exist CC>CD ;But in the Dingyuanhei pig tested group, the genotype effect exist CCCD>DD, and there is no significant difference(P>0.05) between all these three genotype and the four traits associated with the number of born in Wannanhua sows, while in Wannanhei pigs exist significant difference(P<0.05)between the CC,DD genotype effect and the total number born,number born alive in gilts,the number born alive,the number born alive in multiparous sows.
