Wheat(Triticum aestivum L .),one of the most important food crop in China ,is a main source of plant protein as well.With the development of economy,the improvement of living standard and joining WTO,the grain quality of wheat would have been concerned more and more.However,the wheat cultivars which were widely planted nowadays have inferior food processing qualities,especially baking quality.Crossbreeding is one of the most way of quality breeding in common wheat,but it have been limited by the low efficiency and the long period of breeding,etc.Therefore,the development of a more efficient approach to improve grain quality of wheat is of extreme importance.Recent advancement in molecular biotechnology offer new opportunities to improve grain quality of wheat.It has been proved that high molecular weight glutenin subunits were important to wheat flour baking quality .Moreover,genetic transformation of wheat has become well established over the past decade. HMW-GSlBx14+1By15 was discovered in some good quality varieties of wheat,which was important to wheat flour baking quality.In the research, HMW-GSlBx14 gene was transferred into wheat cultivars Shaan Nong 757 via pollen tube pathway in order to improve the grain quality. The main conclusion were as follows: 1.The successful screening of transgenic plants containing HMW-GS lBx14 is based on the establishment of efficient PCR system——gene pool. 2.PCR analysis of 618 transgenic plants proved that 74 plants carried HMW-GSlBx14 gene. 3.Transgenic plant grains identified that by SDS-PAGE showed that HMW-GS lBx14 gene was well expressed in offspring seed(T1) of some transgenic plants.There are four kinds of high molecular weight glutenin subunits: (1)1,7+9,2+12;(2)1,7+9,14,2+4+12;(3)1,14,2+12;(4)1,7+9,14,2+12; 4.Genetic transformation of wheat via pollen tube pathway has been established preliminarily:The best time of insemination is between 30min and 60min, 300ng/μl is the moderate concentration of expression vector.
