In this paper, the apple cultivars of Royal Gala and MATO were explored as the experimental materials to carry on the shoot apex culture and establishment of regeneration system from field shoots and tube shoots of leaf and stem slice. This experiment also discussed many factors that affected regeneration of adventitious shoots, such as genotypes types explants, kinds and levels of hormone, polarity of leaf, shoots age,different culture conditions and methods, etc. Hereby the high-frequency regeneration system of apple was established.The main results were as follows:1. It was found that the explants were the most important factor in the apple aseptic vegetative propagation. Using early spring season developed shoot apex as a source of explants could very easy to get test tube shoot, plants body pollution and changes to avoid and lighten browning maximum outside. Originally and proliferation adopted the LS of medium, hormone composition of originally were BA1.0 mg/L+IBA0.1 mg/L; hormone composition of subculture were BA0.6~0.8mg/L+IBA0.2 mg/L+ GA31.0mg/L.2. Establishment of regeneration system from leaf of field shoots, the culture was with LS +BA5.0 mg/L + IBA 0.3 mg/L had the best result; the regeneration of tube shoots, the culture was with LS +BA6.0mg/L +IBA 0.3mg/L had the best result; the regeneration of stem slices, the culture was with LS+BA5.0mg/L +1BA0.3 mg/L had the best result.3. Regeneration of the stem slice, the culture was with LS +BA6.0mg/L+IBA0.3mg/L has the. best result.4. Regeneration of adventitious bud of different genotype explants, plant hormone added competence to be different.5. The different types of explants: leaf from field shoots .tube shoots and stem slices in case of training the condition to be the same, the regenerated frequency of leaf of field shoots was the highest, more than 70%.6. Induced callus and regeneration of adventitious shoots, must it trains to be secretly earlier stage, complete illumination culture after inoculating, leaf can regenerate too, but dark to is it can improve simple regeneration of adventitious shoots and regenerated frequency of leaf notably to culture. Train for being best to deal with time secretly, simple regeneration of adventitious shoots counted and regenerated frequency reach supreme value three week secretly, it was 6.07 and 72.07%, respectively.7. Leaf polarity test showed regenerated efficiency of back of leaves upward was better than leaf face of leaves upward; and regenerated efficiency was the best from the body leaf midsection.8. The results of shoots age test showed that, tube shoot leaf regenerated better ability of 30-35d after subculture, regeneration of adventitious shoots of simple leaf was counted and regenerated frequency were 6.31, 71.87%, respectively.9. With 1/2LS +lBA0.5mg/L+NAA0.3 mg/L + Agar 6.0g/L + Sugar 15g/L, it had the best result in rooting, rooting rate was more than 80%.
