| In this study, the follicular oocytes of Nanyang Cattle were collected from 28mm follicles, the oocytes of A and B grade were used for in vitro maturation(IVM), in vitro fertilization(IVF) and in vitro culture(IVC). The objective of this paper was conducted to establish an optimum culture system and methods for in vitro produce embryos of Nanyang Cattle, and some affect factors were also investigated. The result as follows:1.Different hormones(E2, FSH+LH+E2, PMSG+hCG+E2 ) were added to maturation medium, the results showed that the maturation rate(MR) and cleavage rate(CR) of FSH+LH+E2 group(72.5 % , 52.6%) were significant higher than E2 group(47.5%,35.4%)(p<0.05). the CR of PMSG+ hCG+E2 group(50.6%) was significant higher than E2 group (p<0.05) .the blastocyst rate(BR) of three groups had no significant differences(p>0.05).2.TCM199 added with 0.6mg/ml BSA or 10%FCS or 10%OCS as maturation medium for IVM. The results showed that the MR of 10%OCS group (73.3%) was significant higher than BSA group(51.7%)(p<0.05). the CR and BR of 10%OCS group and 10%FCS group were all higher than BSA group, but without significant differences(p>0.05).3.Maturation medium added with 0.0%BFF (10%FCS),10%BFF, 20%BFF, 30%BFF respectively for IVM. The results showed that the MR and CR were decreasing with the increasing of BFF concentration. The MR and CR of 10% and 20%BFF group without significant difference when compared with 0.0%BFF(10%FCS) group (p>0.05).,the MR and CR were significant lower than 0.0%BFF(10%FCS) group(p<0.05).4.Oocytes were matured by different maturation time(18h,22h,26h), the MR(52.5%,67.5%,70.0%) were increase when the maturation time prolonged. When matured 22h in vitro, the CR(54.3%) was significant higher than matured 18h(40.0 %)(p<0.05). the BR without any significant difference in three groups(p>0.05).5.Three washing methods(swimming up method, percoll method, direct washing method) were used in this study. The results showed that the CR an BR were without significant difference between swimming up method and direct washing method(p>0.05). the CR and BR of swimming up method(58.3%,25.9%) were all significant higher than direct washing method(47.2%,13.3%)(p<0.05). the CR of percoll method(60.6%) was significant higher than direct washing method(p<0.05), BR was higher than direct washing method ,but without significant difference(p>0.05).6.Different fertilization medium(B0 medium, mTyrode's medium, TCM199) were used on IVF.
