The prevention of vitrification is the key point in rapid propogation of carnation in vitro. The operating construction for shoot tip culture of carnation, and the vitrification in induced culture and subculture have been studied. Different from others, the red-flowering varieties of carnation could not have their vitrification rate to decrease to a low level by adjusting the media and condition of culture, which showed that vitrification is concerned with their genotypes. When cultured in media of Ms+BAO. 2 mg ·L-1+Agar 6.0mg·L-1+Sugar 30g·L-1, pH 5.8-6.0, and under condition of 23°C, photo intensity 2000-4000Lx(10h/d), most varieties of carnation could have their vitrification rates to be controlled less than 10%. The vitrification rates of red-flowering varieties declined as well, but were still high above 60%. Because of the high vitrification rate, the propagation rate of red-flowering varieties only reached 1, which prevented them from rapid propagation and wide application. The technique of tissue culture was used to screen for a clone with low vitrification rate(R clone). After 27 times' subculture, the R clone whose vitrification rate is about 8% and propagation rate reaches 3, has been obtained. The R clone was cultured under different conditions to observe its vitrification character, which showed that its vitrification rate was much lower than that of ck. After 10 times' subculture, the R clone was transplanted in a field, with a result that it kept the traits of its original one and no harmful variation was observed.Data on shoot tip culture reported here may make it feasible for large-scaled propagation of viral -free plantlets of carnation.
