Shoot organogensis culture in vitro and plant regenerated from sprouts explants of Evonymus japonica L. Var. aureovariegatus Reg were studied on this paper. Activities of peroxidase and polyphenol oxidiase in the development of adventitious root were discussed. content of chlorophyll and carotin and anthocyanin in the different culture condition were researchedThe results showed that the best medium for various stages were as follows: (l)Initial medium of stem : MS+BA4.0 mg/L+TDZ0.05 ~ 0.1 mg/L+IBA0.1 mg/L+NAA0.01 mg/L + 3%sucrose; Initial medium of laminae: MS+IBA0.1 mg/L++TDZ0.05 mg/L; Draw materials season: April.(2) Clump shoots genesis from callus proliferaten.medium : MS+BA1.0 mg/L+NAA0.1 mg/L+IBA0.5 mg/L+3%sucrose; Clump shoots genesis from axillary buds proliferate nmedium : MS+BA2.0mg/L+NAA0.1mg/L+IBA0.01mg/L+ 3%sucrose; Succeed era period: 30d;Succeed era algebra: four era.(3) Rooting medium: 1/2MS+IBA0.5 mg/L+ NAA0.1 mg/L +1.5%sucrose;Rooting Of outside cuvette: 200mg/L IBA+200mg/L NAA.(4) Forging seeding transplant: vermiculite 25%+perlite25%+riversand50%(5) Peroxidase activity was high in the root formation of inner Test-tube rooting than out Test-tube rooting, peroxidase activity increased in the root formation of inner Test-tube rooting than out Test-tube rooting and peroxidase activity increased and then decreased in the root formation of out Test-tube rooting. Polyphenol oxidiase activity decreased and then increased in the root formation of inner Test-tube rooting and out Test-tube rooting.(6) Content of chlorophylla was high and little of effect in different culturecondition. Content of chlorophyllb and carotin was low in different culture condition and they had prominent effect in illumination hour. Content of anthocyanin was zero in different culture condition.
