This paper mainly dealt with the study on shoot organogensis culture in vitro and plant regenerated from sprouts explants of Elaeocarpus sylvestris (Lour.) Poir and study on activities of peroxidase and polyphenol oxidiase and content of endogenous 1AA and ABA in the development of adventitious root.The results showed that the best medium for various stages were as follows: (l)Initial medium: MS+BA1.0-2.0 mg . L-1+NAA0.05 mg . L-1+3%sucrose; (2) Clump shoots genesis from callus proliferaten.medium: MS+BA1.0 mg L-1+NAA0.5 mg L-1+ 3 % sucrose; Clump shoots genesis from axillary buds proliferaten.medium: MS+BA2.0 mg . L-1+IBA0.1 mg L-1+ 3% sucrose; (3)Efficiency seedling induction medium: MS+BA0.5 mg L-1+IBA0.1 mg . L-1+GA1.0mg L-1+3% sucrose; (4)Rooting medium: 1/2MS+IBA3.0 mg L-1+2%sucrose. In root induction culture, the rate of root formation with membrane filter is higher than that of root formation with common membrane.The results of activities of peroxidase and polyphenol oxidiase and content of endogenous IAA and ABA in the development of adventitious root were as follows:(5) After treated with IBA or NAA, the rate of root formation is high. From zero day to the fourth day of adventitious root development, peroxidase activity decreased and then increased. When roots began to elongate, peroxidase activity decreased. After treated without hormones, the rate of root formation is zero, In zero day to sixteen days of adventitious root development, peroxidase activity increased and then decreased. Polyphenol oxidiase activity is very low during adventitious root development.(6) After treated with IBA and NAA, the content of endogenous IAA and ABA increased. When the root can be seen, the content of endogenous IAA and ABA began to decrease. Treated without hormones, the content of endogenous IAA was always low; thecontent of endogenous ABA increased constantly until the root formation and then decreased. Treated with IBA and NAA,, the rate of IAA to ABA is 2.5~3. 2 and treated without hormones, the rate of IAA to ABA is lower than 1.2...
