The pyrimidinylbenzylamine herbicides are a series of strong herbicidal activity compounds developed in recent years with Chinese own autonomous intellectual property. They are conjectured as a kind of pre-herbicides, i.e. the pre-herbicides are metabolized and bioactived to the compounds that inhibiting ALS activity after it is absorbed into the sensitivity plants, according to the preliminary study of ZJ0273, a pyrimidinylbenzylamine compound. ZJ0777 was synthesised recently at Zhejiang university and belonged to the pyrimidinylbenzylamine compound. In this paper, the effect of ZJ0777 on the upgrowth and POD, EST, ATPase isoenzyme of Sorghum (sensitivity plants)and Brassica (resistent plants ), and the ZJ0777 metabolism with POD in vitro were studied.1. The erminating Sorghum and Brassica seeds were treated with different concentration of ZJ0777, it was found that 2 ppm of ZJ0777 couLd markedly inhibits the growth of Sorghum roots and stems, but no effect on Brassica roots and stems. However, after handled with Sorghum extraction, 2ppm ZJ0777 couLd inhibiting the growth of roots and stems of Brassica. The resuLts showed that ZJ0777 is possibly translated into poisonous compounds under the action of Sorghum crude POD extraction.2. The POD activity of Sorghum and Brassica seedlings (2~3cm) was detected, the resuLts showed the POD activity of Sorghum seedlings was higher than that of Brassica seedlings. Then the seedlings were treated with 2 ppm ZJ0777 and their POD activity were detected in 6h, 12, and 24h. The resuLts showed the variety of POD activity have significantly difference between Sorghum and Brassica: the Sorghum seedlings POD activity gradually declined in 24h, oppositely did in Brassica. Besides, the pattens of POD, EST, ATPase isoenzymes in 6h, 12,and 24h were analysed.3. The crude POD extraction of Sorghum and Brassica seedlings were electrophresis in LDS-PAGE, the POD active strips, detected by dyeing ofHAC-Benzidine, was cut and soaked in phosphate sodium buffer (O.IM pH7.4) containing 2ppm ZJ0777 overnight, a new compound was detected in HPLC (C18-ODS column).This indicated that POD can cause the change of ZJ0777 configuration and thereout we presume POD is concerned with the ZJ0777 metabolism in the plant.
