Studies Of The Host Range Of Tomato Yellow Leaf Curl China Virus And Its Satellite DNAβ And Cross-protection

The host range of Tomato yellow leaf curl china virus (TYLCCNV) Y10 isolate (Y10) with their associated DNAβ ( Y10β) were determined. The stability of Y10β in hosts and cross-protection mediated by a βC1 deletion Y10β were also investidated.PCR detection showed Y10 and Y10β could systemically infect Nicotiana benthamiana, N. glutinosa, N. tobacum, Petunia hybrida, Lycopersicon esculentum and Phaseolus vulgar is, but could not infect Glycine max. It was found that YIO and Y10β could replicate for about 1-2 weeks in P. vulgaris, which is different from other hosts. Y10 and Y10β could not be detected by PCR or TAS-ELISA in G max. Y10 and Y10β can induce typical symptoms such as downward leaf curling, stunt, vein thickening, enation in N. benthamiana, N. glutinosa, N. tabacum and L. esculentum. Typical symptoms except enation were also found in P. hybrida. In P. vulgaris, vein thickening were observed. Y10 and Y10β could replicate stably for a long time in N. benthamiana, N. glutinosa and N. benthamiana, they could be deteced in plants 210 days post inoculation.Pre-inoculation of βC1 gene detective mutumant of Y10β (Y10 Δ C1β) with cognate virus could mediated cross-protection to challenge-infection of cognate helper viruses and associated DNAp. All N. benthamiana and N. glutinosa plants pre-inoculated with Y10 and Y10 Δ C1β exhibited mild symptoms after challenge inoculation with Y10 and Y10β, but protection effects in N. benthamiana were better than in N. glutinosa, and protection effects against cognate DNAβ were better than DNAP associated with Tobacco curly shoot virus (TbCSV). Y10ΔClβ could not mediated cross-protection to challenge-infection of Y10 and Y10β or TbCSV and its DNAβ in L. esculentum.
