Low temperature does harm to survival for lots of plants severely, the loss of crops caused by lowtemperature is up to hundreds of billions RMB in the world every year. Studying and improving thecold tolerance of plants play a significant role in the theoretical and actual aspects. With the increasingof people's number and decreasing of infield area, people gradually pay more attention to improvingthe yields of crops, but the climate condition is one of important factors for affecting crops production.Cold damage always limit the increasing of crops productivity, severe cold damage even causes cropsto die out. If we would cultivate crops breed with cold tolerance, it is probable of enlonging the sterileperiod of crops, which prevents from bad affects caused by abrupt low temperature and plays importantroles in improving the crops production. It is the best way that crops breed with cold tolerance arecultivated. It is difficult to gain good breed by traditional methods because of the complication ofstresses tolerance character of plants and the complexity between it and the site chain relations ofquantitative character. With the development of biology technique, the stress tolerance by transformingforeign genes has become dominant methods for improving modern plants breed. In recent years, theresearch on transgenic plants has made great advancement, people has obtained over one hundred kindsof transgenic plants related to more than fifty species. At present, people has cloned cold toleranceICE1 by Arabidopsis, constructed plants expressing vectors and transformed Arabidopsis. It has beenincreased for cold tolerance positive Arabidopsis plants. This research cloned cold tolerance gene ICE1from Arabidopsis's RNA and cloned cold inducible promoter rd29A from Arabidopsis's genome DNA,connected CaMV35S promoter and cold inducible promoter rd29A with ICE1 respectively andconstructed the plant expressing vectors pBin35SICE1 and pBinrd29ICE1. This research transformedthe tobacco with these 2 vectors and getted over 100 resistance plants. PCR identification showed thatabout 44% were positive. This research contributes a new and efficient pathway to the cold tolerance intobacco.
