Construction Of PSY Plant Expression Vector And Transformation Into Eustoma Grandiflorum

PSY (Phytoene synthase) is one of the key enzyme during the course of Carotenoids biosynthesis. The biosynthesis of carotenoids can be facilitated through the transformation of PSY. Carotenoids concludes almost important pigment from red to yellow, which is used in genetic engineering of flower color. Eustoma grandiflorum is one of the pot flowers and cut flowers. It is a more effective and quicker method than conventional breeding to alter flower colour using genetic engineering.The plant expression vector of pBI121-PSY was constructed in the research. Through carrying out of the system of bud regenration from the leaf of Eustoma grandiflorum, the studying of the factors affecting transformation. The results were as follows:(1) The PSY from medler was cloned in plant expression vector pBI121. The vector was transformed into Agrobacterium tumefaciens LBA4404, that is to say, the plant transformation engineering strain was constructed.(2) The system of bud regenration from the leaf of Eustoma grandiflorum was carried out.The results indicated that the optimal culture midium for adventitous bud differention was MS + 6-BA1.0 mg/L + NAA 0.15 mg/L, the optimal culture midium for adventitous bud growth was MS + 6-BA 0.5 mg/L + GA₃ 0.6 mg/L, the optimal culture midium for root induction was1/2 MS+ IBA 0.5 mg/L.(3) The fit concentration of Cef to inhibit the growth of Agrobacterium tumefaciens LBA4404 was 400 mg/L.
