| A series of defense responses may take place when the plant is treated with some compounds, salt and pathogens, such as fungi, bacterium and virus, or any other stresses, usually accompany with the generation of some new proteins, named pathogenesis-related proteins(PRP). According to the latest researches, PRs can be classified into 17 classes, such as PR1, PR2, PR3, and PR4. Despite the anti-fungi function of PR1, the biological functions of other PRs was frequently reported, including salt tolerance and virus resistance. It is becoming more and more popular that researching the function of different PR genes, especially in plant defense by transgenic technology. In this study, we cloned tobacco PR1b gene (NCPR1b), PR4a gene (NCPR4a), and rice RSOsPR10 gene (RSOsPR10), and transferred corresponding expression vectors to Nicotiana tabacum NC89 and Nicotiana tabacum L.cv Samsun, respectively, by Agrobacterium tumefaciens-mediated transformation. A series of transgenic plants was obtained. We performed salt-tolerance and anti-pathogen analysis of the T1 progeny. The main results are as follows:1. According to the reported gene sequence, three genes were cloned by RT-PCR technique using specific primers, which are NCPR1b form NC89 with 506 nucleotides in the cDNA, NCPR4a also from NC89 with 460 nucleotides in the cDNA, and RSOsPR10 form rice ,whose cDNA contains 586 nucleotides.2. Five different plant expression vectors were constructed, such as pBI-Z-PR1b containing sense NCPR1b gene, pBI-F-PR1b containing antisense NCPR1b gene, pMR-PR1b with a MAR enhancing expression and pBI-PR4a and pROKâ…¡-PR10 containing sense PR4a and sense RSOsPR10, respectively.3. Using the NC89 as the receptor, the pBI-Z-PR1b and pBI-PR4a were introduced by Agrobacterium tumefaciens-mediated... |
