Chilling and freezing injury was one of the common environment forces factor which can cause significant loss in plant productivity and limit growth areas of plants. Hence how to improve the cold resistance of plants was always the scientists' ideal and the pursue. The separation, the identification, the cloning and the transformation of cold-resistant genes always was one of hot spots in the domestic and foreign scientists studies field. Recently, along with the great advances in the researches of molecular mechanisms of cold injury, many cold-resistant genes have been found, including all kinds of regulatory genes and functional genes. Experiencing the conventional breeding method was very difficult to improve the plants cold resistance, the studies of genetic engineering for cold resistance in plants have also been carried out extensively in the past few years. Genetic engineering of cold- resistant plants had made a great progress at home and abroard.The gene kin1 separated from Arabidopsis thialiana encoding 6.5kD poly-peptide. It had an unusual amino acid composition being rich in alanine, glycine and lysine. Northern blot analysis revealed that the level of kin1 mRNA was increased 20-fold in cold-treated plants. In addition to being expressed in cold, kin1 was also induced by water stress and the plant hormone abscisic acid (ABA), which has been suggested to be a common mediator for osmotic stress responses and cold acclimation in plants. Sequence comparisons showed that the kin1 gene product had similarities to fish antifreeze proteins (AFPs). The gene product can enhance cold-resistance of plants. In this paper the cDNA of Arabidopsis antifreezing gene kin1 was amplified from its total RNA by RT-PCR method , then we constructed its expression vector for the forther use to be transfered into plants in the future.C.microcarpa was one of the important ornamental fruit trees. It was so cold in the north so few quantities of potted C.microcarpa can only be found in the room. Cultivating the cold resistant C.microcarpa, without doubt had important economic benefit and ecology value. A new method was provided...
