In this passage we studied the dissolution of insoluble phosphate and the conlonization capability in soil and plants of Klebsiella terrigena 102.The results showed that 102 has better solubilizations to Ca-P,Al-P and RP.102 was inoculated to plate media content with Ca3(PO4)2 and the diameter of transparent is 59.5mm.When the liquid cultures added with KH2PO4 in the mount of 0mgP.L-1, 10 mgP.L-1, 20 mgP.L-1,respectively,it showed that 102 can grow well and the dissloved phosphate were reach to 72.83 mgP.L-1, 61.92 mgP.L-1,66.36 mgP.L-1 in the second day,the pH of the culture medium decreased and a great deal of organic acid were produced.The analysis of HPLC indicated that the organic acids in liquid culture were mainly acetic acid and lactic acid.Our study revealed that 102 strain had a significant eddect on the dissolution of insoluble phosphate in Magantu and fluvo-aquic soils.By inoculated with 102,the content of available P increased 11.89mg.kg-1 and 9.90mg.kg-1 respectively in sterilized Magantu and fluvo-aquic soil,compared with CK.Three inoculation methods(seed-soaking,mixed with soil and root-soaking) were conducted. We found that 102 marked with GFP had the highest colonization in tomato rhizosphere by the mixing-with-soil,and its colonies reached to 106cfu/g in the 4th week.Confocal laser scanning microscope analyses reveal that 102 can colonize in ryegrass.The colonization capacity can reach to 107 cfu/g fresh weigh in Pennisetum anerlcanum or 106cfu/g fresh weigh in ryegrass.102 colonization capability in soil was tested by addition with Magantu soil extract,fluvo-aquic soil extract,CGF from chlorella,antibiotic and compost extract in liquid culture,respectively.It was proved that fluco-aquic soil extract,CGF from chlorella,antibiotic and compost extract in liquid culture could increase 102 colonization in fluvo-aquic soil,and compost could sigificantly improve 102 colonization in Magantu soil.
