Banana panama disease [Fusarium oxysporum f.sp. cubense (E. F. Smith) Snyder] is an important quarantine plant disease. Fourteen strains of Fusarium oxysporum f.sp. cubense were isolated from banana plantations of Xiangcheng district , Changtai county , Zhangpu county and Pinghe county of Zhangzhou . FOCAAA4 and FOCAAA9 from Musa (AAA) , FOCABB1 from Musa (ABB) were inoculated separately in banana cultivars of Musa (AAA) Taiwan, Brazilian, red and a cultivar of Musa (ABB) , FOCAAA4 and F0CAAA9 can infect Taiwan , Brazilian, red and Musa (ABB), FOCABB1 only infected the Musa (ABB). FOCAAA9 and FOCABB1 were inoculated on PSA medium , CMA medium , pseudostem Musa (AAA) medium and pseudostem Musa (ABB) medium respectively, The results showed that all are different in colony characteristic , growth speed , spore shape , spore yield , spore types. Using vegetative compatibility to test fourteen strains of Fusarium oxysporum f.sp. cubense, result indicate twelve strains of Fusarium oxysporum f.sp. cubense from Musa (AAA) belong to VCG1 , two strains of Fusarium oxysporum f.sp. cubense from Musa (ABB) belong to VCG2 . A polymerase chain reaction (PCR) protocol was developed to amplify internally transcribed spacer (ITS) region of ribosomal DNA of F0CAAA9 and FOCABB1 , and the generic PCR products were sequenced and analyzed.Result indicates the phylogenetic relationship between FOCAAA9 and FOCABB1 is very similar, comparing their ITS sequences, the homology of ITS1 is 98.6% and the homology of ITS2 is 98.0% . As above, we think FOCABB1 from Musa (ABB) is Fusarium oxysporum f.sp. cubense race1, FOCAAA4 and FOCAAA9 from Musa (AAA) is Fusarium oxysporum f.sp. cubense race 4.The methods of vegetative compatibility and PCR were used to detect pathogenic fungi.The results showed that vegetative compatibility test can identify the race of Fusarium oxysporum f.sp. cubense in Fujian, PCR method can detect fungi in banana tissue rapidily.
