Banana Fusarium wilt is an important banana vascular system disease,the pathogen of which is Fusarium oxysporum f.sp.cubense(Foc).Foc1 and Foc4 are the main races in China of 4 races.Foc1 mainly infects Fenjiao(AAB)and can not infect Xiangyajiao(AAA),but the reasons for the difference of different kinds of bananas by Foc1 are not clear at present.However,the reason of the difference in Foc1 infection among different types of bananas is not clear.Secretory protein is an important pathogenic factor of Foc and plays an important role in the process of infection.1.Six kinds of medium suitable for Foc1 conidia germination and secretion proteome analysis were screened.The results showed that Foc1 conidia had a high germination rate in NCM-4A medium(10 g glucose,4 g aspartic acid,20 x nitrate 50 mL,1000 x vitamin 1 mL,1000 x trace element 1 mL,add deionized water to be 1 L,pH=6.5),which did not contain the source of nitrogen,was beneficial to the study of secretory protein by Foc1.2.The critical period of bud tube elongation(7 h)was selected as the research object.The Foc1 secretory protein was extracted by acetone precipitation method induced by fenjiao and Brazilian banana tissue.The results showed that there were 124 secretory proteins in Foc1 after induction,of which 49 were classical secretory proteins(39.5%),18 were non classical secretory proteins(14.5%)and 57 secretory proteins(46%)were unknown secretory pathways.3.Compared with the control,64 secretory protein differential expressed was induced by Fenjiao.(1)There were 31 secretory proteins expressed only in the condition of Fenjiao,20 secretory proteins were expressed only in the control,and 13 secretory proteins were differentially expressed after the induction of Fenjiao.(2)Using the software of SignalP and other bioinformatics software to predict and analyze 64 differentially expressed secretory proteins.The results showed that there were 28 classical secretory proteins and 7 nonclassical secretory proteins.(3)Using the software of dbCAN to predict and analyze the CAZymes.The results showed that there were 7 cell wall degrading enzymes(CWDEs).(4)GO enrichment analysis showed that the function of secretory proteins was mainly catalytic activity,binding,structural molecule activity,molecular function regulator and signal transducer activity;KEGG enrichment analysis showed that these secreted proteins were mainly involved in metabolic process,cellular process,cellular component organization or biogenesis,biological regulation,and regulation of biological process;There are 1 pathways that are significantly enriched,and pathways such as valine,leucine and isoleucine biosynthesis are significantly affected.4.Compared with the control,90 secretory protein differential expressed was induced by Brazilian banana.(1)There were 32 secretory proteins expressed only in the condition of Brazilian banana,42 secretory proteins were expressed only in the control,and 16 secretory proteins were differentially expressed after the induction of Brazilian banana.(2)Using the software of SignalP and other bioinformatics software to predict and analyze 90 differentially expressed secretory proteins.The results showed that there were 34 classical secretory proteins and 12 nonclassical secretory proteins.(3)Using the software of dbCAN to predict and analyze the CAZymes.The results showed that there were 4 cell wall degrading enzymes(CWDEs).(4)GO enrichment analysis showed that the function of secretory proteins was mainly catalytic activity,binding,structural molecule activity,antioxidant activity and molecular function regulator;KEGG enrichment analysis showed that these secreted proteins were mainly involved in metabolic process,cellular process,cellular component organization or biogenesis,biological regulation,and response to stimulus.5.Compared with the Fenjiao,34 secretory protein differential expressed was induced by Brazilian banana.(1)There were 5 secretory proteins expressed only in the condition of Brazilian banana,22 secretory proteins were expressed only in the condition of Fenjiao,and 7 secretory proteins were differentially expressed after the induction of Brazilian banana.(2)Using the software of SignalP and other bioinformatics software to predict and analyze 34 differentially expressed secretory proteins.The results showed that there were 17 classical secretory proteins and 5 nonclassical secretory proteins.(3)Using the software of dbCAN to predict and analyze the CAZymes.The results showed that there was 1 cell wall degrading enzymes(CWDEs).(4)GO enrichment analysis showed that the function of secretory proteins was mainly catalytic activity,binding,structural molecule activity and antioxidant activity;KEGG enrichment analysis showed that these secreted proteins were mainly involved in metabolic process,cellular process,response to stimulus,cellular component organization or biogenesis and regulation of biological process;There are 1 pathways that are significantly enriched,the pathway synthesis of secondary metabolites of was significantly affected.
