There are lots of cultivars of tea in china. The genetic diversity and relationship of thirty-nine varieties of tea plants in china was studied based on RAPD and ISSR molecular markers.The results are as follows:l.Based on the previous research, according to traits of tea,the method about extraction and purification of tea genetic DNA are simpe and rapid and accurate.2.According to the require of experiment,comparing the common procedures of molecular markers of RAPD and ISSR,some modifications had been adjusted.The result was to set up the reaction system of RAPD and ISSR fitting for analyzing the resources of tea.3.Twenty primers of 10 base pair were selected out from 100 random primers and were used to analyze with RAPD among 39 accessions, which generated 196 bands,of which polymozphic bands number was 171, the percentage of polymorphism equaled 87.2%; 15 primers, which could amplified clearly bands, were selected out from 40 ISSR primers. As a result, 143 bands were generated, of which polymorphic bands number was 131, the percentage of polymorphism was 91.6%.4.There was a high correlation between RAPD and ISSR (r=0.912, n=741), indicating that the two methods were highly consistant and suitable for detecting genetic diversity and genetic relationships in tea. But some cultivars still clustered different between the two makers.5.According to the clustering analysis of RAPD results, it proved that the genetic similarity index was 0.17—0.97. According to the clustering analysis of RAPD results, it proved that the genetic similarity index was 0.21—0.95.From the result, genetic diversity and similarity and genetic relationships were revealed.6.The genetic map of thirty-nine varieties of tea were constructed.The tea cultivars can be clearly distinguished by the genetic map of RAPD and ISSR.The relationship of tea cultivars were validated by cluster analysis .
