Soybean (Glycine max L. Merr.) as one of the most important resources of oil and protein is widely planted in China As the fast increase of population and decrease of plantation, the exploration of saline soil would be very important and efficient, which has become one of the hot spots nowadays.Soil salinity is one of chief environmental factors that limit the crop yield potential. Soybean is middle sensitive to saline, and its salt tolerance capacity is very low so that it is really important to breed soybean varieties with high salt tolerance. Due to lack of good varieties with different excellent characteristics, it is hard for researchers to breed soybean varieties with high salt tolerance by using traditional breeding method whose process is relatively slower than transgenic methods. With the development of molecular biotechnology, it is efficient to improve different characteristics of soybean. In this experiment, cotyledon nodes of general soybean were used as receptor materials. Studies have been done on genetic transformation of salt tolerance genes Betaine aldehyde dehydrogenase Gene (BADH) and Choline monooxygenase Gene (CMO), which make a good foundation for breeding varieties with good capacity to endure saline stress. The sensitive characteristic on salt of soybean cotyledon nodes and soybean seedlings were studied. Critical concentrations of sodium that would be very useful for further selection and research were determined respectively for both soybean cotyledon nodes and soybean seedlings. The physiological and biochemical indexes of transformed soybean seedling and control under the same sodium concentration were detected in this experiment in order to determine the salt tolerance capacity of transformed soybean. The results of studies were acquired as follows:1. Research on genetic transformation methods of soybean(1) Research on transformation of salt tolerance gene into cotyledon node of soybean by Agrobacterium tumefaciens-mediated MethodThe cotyledon nodes of Jinong13 and Jinong17 were chosen as experimental materials to transform salt tolerance genes BADH and CMO by using Agrobacterium tumefaciens-mediated Method. Researches on the conditions of transformation such as different time of incursion, the concentration of...
