Pine tree is one of the major trees in Sichuan province, and large areas of pine trees are appeared both in natural and artificial forests. Recently, the needle cast disease has occurred wildly in Sichuan forest region, and severely threatened pine trees' growth in some area. The authors had an investigation on this disease in major pines tree areas, and found that Lophodermium spp. was the pathogenic fungi which caused the needle cast disease. Various species in this genus have been identified, and their culture characteristics and their genetic diversities were analyzed.More than 40 samples collected from main pine forest areas in Sichuan province (Yaan Mingshan, Ganzi Luding, Ganzi Kangding, Meishan Danling,Chengdu Pujiang, Neijiang Zizhong, Zigong, Yibin, Luzhou, Nanchong, Dazhou,Xichang Mianning) were sorted, cultured, and identified by their morphologic characteristics to species. Based on the classification standard proposed by Minter, seven species of Lophodermium genus were identified They were L. conigenum (Brunaud) Hilitz, L. pinastri (Schrad) Chev, L.sichuanense Qiu et Liu, L. indianum Singh et Minter, L.australe Dearn, L. erlangshanense (temporary name) , and L. kumaunicum (Minter et M.P.Sharma). Among them, L. erlangshanense is a new species and L. kumaunicum is a new record in Sichuan. Full-blown samples were isolated in PDA culture media by ascocarp isolation techniques, and 24 strains were obtained. Their culture characteristics were recorded and data were used for genetic diversity analysis.Genomic DNA from 24 strains of Lophodermium were extracted by the improved method of precipitation, and Random Amplified Polymorhpic DNA (RAPD) was conducted.
