A regeneration system of tissue culture in vitro using stem with bud and radicle as explant was established on Dioscorea nipponica Makino and experiment with polydesign, The main results can be summarized as follows: 1. Inducement of callus: The explant treated with 4℃can enhance the frequency of Inducement of callus, which can be improved from 9%-13% compared with CK. The damnify will be produced if explant is leaved in low temperature excess six days, which will affect the ability of inducing callus. Different explants have different frequency of inducing callus, the frequency of the stem is the highest and which can be reach 75% on the culture medium with 6-BA1.0mg/L+2,4-D4.0 mg/L; the radicle takes second place and which can be reach 72.5%, but the callus induced by radicle proliferate quickly compared with callus induced by stem.2. Inducement and proliferation of buds: The gemma can be induced when the callus was transferred into the culture medium with MS+6-BA2.0mg/L+NAA2.0mg/L and the highest inducement frequency can be reach 75.5%. The concentration of 6-BA between 1.5-2.5mg/L is optimal. The gemma can be induced from axilla bud on the culture medium with 6-BA2.0mg/L+NAA2.0mg/L+AC0.5g/L and the highest inducement frequency can be reach 85.7%. The optimal culture medium of proliferation of buds is MS+6-BA 1.5mg/L +NAA0.5mg/L and the proliferation coefficient can be reach 3.22.
