Dioscorea nipponica Makino is a plant of the family Dioscoreaceae, which is distributed in Inner Mongolia, Heilongjiang, Jilin, Liaoning and other provinces in China, one of the main sources is diosgenin. The plant is not only a main resource to medicines for cardiovascular diseases, but also is a widely used raw material for the production of steroid and contraceptive.The purpose of this study was to make research on seeds dormancy and germination of Dioscorea nipponica Makino and general coverage of the pine needles in agricultural production on the impact of seed germination of Dioscorea nipponica Makino and microbial area by the research of allelopathy, we could improve and optimize the cultivation techniques of Dioscorea nipponica Makino.The intrinsic inhibitory substance existed in Dioscorea nipponica Makino seeds, which strongly inhibited the growth of young root of than Dioscorea nipponica Makino seed germination, and inhibited the content of chlorophyll, the fresh weight and the activeness of a-amylase of wheat than seed, germination, and also significantly inhibited the germination of Dioscorea nipponica Makino seeds. The seeds treated with ether twenty-four hours in the constant temperature could increase the germination percentage. The research results of the seed dormancy showed, the seeds of Dioscorea nipponica Makino reached highest germination rate (76.5%) at the temperature treatment in alteration of 15℃and 25℃, and soil was fertile sandy.Seed germination had been increasingly suppressed with the increasing amount of pine needles solution in the process of seed germination. Three fungal microorganisms (bacteria, actinomycetes, mildew) had been varied in the different degrees of inhibition with the increasing concentration. Thus we proved that Dioscorea nipponica Makino should use pine needles covering water to prevent soil crusts in nursery production, this method affected seed germination and reduced the quantity of microorganisms of seedbed, that is to say, the pine needle extracts and the steep had impact on seed germination and soil microbial area. So we obtained that there is the existence of allelopathy among them.The determined results and statistical analysis results of the samples growth in different growth years showed that the contents of diosgenin highest in sample growth in third year, and the determined results and statistical analysis results of the samples collected from spring to autumn showed that the statistical analysis results of sample collected in May 20 were best.
