The subject of the study is to supply a plant sterilization with effective parts. It is significant for producing the Radix gentianae of GAP standard, and the study of harmless pesticide.By applying the methods of chemistry and pharmcodynamic, the study on screening the antifungal activity parts against Septoria microspra Speg. for Rhizoma anemarrhenae. have been completed. The total saponins of Rhizoma anemarrhenae. were proved to be the best active antifungal parts, and the minimum inhibitory concentration was 0.02g/ml. Conditions for the recovery were studied by orthogonal design and UV-spectral method, guided by the content of Sarsasapogenin. The best extraction process was determinded to be three refiuxs with five times of 75% alcohol for two hours.The effective constituents were accumulated by using the content of Sarsasapogenin as index. Firstly, AB-8 macropore absorbed resin was choosed as the optimal technique for isolation, and the concentration of sample was 0.5g/ml, the flowing speed of absorption ang unabsorption were 1.0g/ml and 2.0g/ml. Secondly, the parameters in the concentration and volume of the elution were investigated mainly. The technique of preparation was established as following: elute with water (5BV) elute with 40% ethanol (7BV). The content of the total saponins of Rhizoma anemarrhenae. in large experiments were 66.48%. The preparation of the effective constituents would contribute to the experiment of pharmcodynamics later.By using the content of Sarsasapogenin as index, and the inspectings of precision, stability, repeat, and the average recovery, The method of determination of content for Total saponins of Rhizoma anemarrhenae. was established. By doing the inhibition experiment, The total saponins of Rhizoma anemarrhenae. were again proved to be the best antifungalactive parts, and the minimum inhibitory concentration was 0.02g/ml. The conclusion would supply the basis for production and studying later.
