Acacia melanoxylon is one of the main timber tree species in South China which is imported in recent years.It has good adaptation,high yield, and many other uses.In this experiment,chose 4 provenances of blackwood's seeds as basic material,researched about every steps of seeds axenic bourgeon, proliferation,rooting induction,transplantation etcAnalysed the key factors of proliferation and rooting ;Seeds's hypocotyl. were used as the initial material for the inducement of calli, 4 provenances blackwood's calli systems were established ,and polarized seedling from calli; and then the preliminary study of cell suspension culture on blackwood.The main results were described as follows:The establishment of blackwood axenic system. Blackwood's seed capsule was rigid and compact,deep in oil of vitriol for 10 minutes,reached 86.7% burgeon rate; sanitized by 0.1% HgCl2 for 15 minutes,inoculated to the medium of MS supplemented with AC1.0 g/L,kept vigorous growth .The proliferation of blackwood axenic seedlings. As far as the influence of different plant growth hormones was considerd.the medium on the proliferation screened out with orthogonal design was, MS supplemented withl.O mg/L 6-BA, KT1.0 mg/L and 0.4 mg/L IBA.the proliferation could reach 3.15 by 30 days ;in the second and repetitious cultivation,MS medium supplemented with 0.4 mg/L KT, 0.4 mg/L IBA and 20g/L sugar was the best;So we chose MS medium supplemented with 0.4 mg/L IBA and 15 g/L sugar to keep the plantet strong,kept the blackwood persistent and rapid proliferation.Rooting and transplantation of blackwood.Thinked about the influence of different kind and concentration hormones on the axenic seedlings rooting rate, screened out the medium was 1/2MS supplemented with 0.4 mg/L IBA, 0.2 mg/L IAA and 15 g/L sugar; the transplantation medium was ocher: turf=2:1. It was very important to keep the balance of leaves moisture within a week after transplantation for developing the survival rate.And in this way, the survival rate could be up to 80%.The establishment of blackwood calli system.Chose four provenances of blackwood's seeds's hypocotyl as basic material for the inducement of calli,under the light,consider the influence by diffenent kind and concentration of hormones and accretion, the MS medium supplemented with 1.0 mg/L 2,4-D, 2.0 mg/L BA was the best;and the proliferation medium were MS medium supplemented with 0.5 mg/L 2,4-D, 2.0 mg/L BA and 300 mg/L LH and B5 medium supplemented with 2.0 mg/L BA, 1.0 mg/L KT, 1.0 mg/L NAA and 300 mg/L LH.could keep proliferating for more than one year.The regeneration of blackwood calli.Under the light,considering the influence of plant growth hormones and accretion. MS medium supplemented with, 1.0 mg/L BA 1.0 mg/L KT, 0.5 mg/L NAA and 300 mg/L LH,the calli of 19495 provenanc could reach 30% differentiation rate; MS medium supplemented with, 1.0...
