As a member of Cucurbiteae, biter melon is an annual trailing plant. The fruit of biter melon contents many kinds of essential amino acid and microelement which have high nutritive value. Furthermore the active substance of bitter melon which also have various medical functions, such as decreasing blood sugar, antibiotic, antitumous and enhancing the body immunity. So as not onl -y a vegetable crop but also an important medical plant, bitter melon has a high studing value. The experiment carried on systematic research to Nanyu bitter melon tissue culture system; optimized Agrobacterium mediated genetic transformation system of Nanyu bitter melon, and the transgenic plants study were obtained. Main result and conclusion were the following:1 The foundation of bitter melon tissue culture system1) The seed would tend to carry bacterium after a long time preservation. The aseptic germin -ation was the best in the 1/2MS medium.2) Multiple buds can regenerated directly from cotyledonary node, and it was extremely diff -ictult to obtain multiple buds from leaf, stem segments and callus.3) A direct high frepuency regeneration system of bitter melon was established successfully, which used cotyledonary nodes as explants. Using the MS medium as the basic medium, optimum multiple buds regeneration medium for cotyledonary nodes section was supplemented with 3.0 mg·L-16-BA and 0.2 mg·L-1 IAA, giving a regeneration frepuency of 93.8 %.4) When the cotyledon of sterile seedlings was light green, the cotyledonary nodes were most suitable to induce the multiple buds directly.5) Although some differences were present among genotypes,a certain of concentration 6-BA and IAA matching were favorable to induction multiple buds, but higt concentration 6-BA was not fav -orable to elongation.6) Adventitious root can be obtained easily from regenerated plants of bitter melon. The low concentration of IAA(0.2 mg·L-1) in 1/2 MS medium can induce normal adventitious roots, wile the result of NAA was not good.2 The optimization of Agrobacterium tumefaciens-mediated genetic transformation of bitter melon.1) The bitter melon cotyledon nodes was insensitive to kanamycin,and 100 mg·L-1was the suitable screening concentration,the multiple buds inducing of cotyledon nodes were inhibited obviously.
