The Brassica juncea chitinase gene BjCHI1 is induced by wounding, treatment with jasmonate (JA), insect feeding or by fungal infection; indicating the promoter of BjCHI1 is an inducible promoter. Currently, constitutive promoters were used in almost all reported transgenic plants. However, due to their constitutive expression pattern, exogenous genes can express in all tissues of plants at any development stage, which would lead to greatly waste of both materials and energy and increase negative influences on metabolism of host plants. Therefore, it is very important to analysis tissue-specific promoters and inducible promoters. In this study, a predicted inducible promoter from Brassica juncea was transferred into tobacco, and its function was analysised.A fagement (280bp) was forwarded from 5' BjCHI1 promoter BjC-p by adaptor-PCR, and obtained the promoter fragement BjC-pl(1378bp). This promoter is not registered in GenBank.The plant expression vector (pBICHPO) harboring the fusion gene BjC-p0 :: GUS was constructed. By Agrobacterium tumefaciens - mediated, three deletion constructs including pBICHPO, pBICHP2 and pBICHP5 were transferred into tobacco K326. 103 positive transgenic tobaccos were obtained, 50 plants were detected by GUS gene (29 with pBICHPO, 21 with pBICHP2 and 0 with pBICHP5).When the transgenic tobaccos with 4 leaves, there was hardly any GUS activity being tested in pBICHPO and pBICHP2 transgenic tobaccos under no...
