Objective:Establish different aged postmenopausal model and to explore the impact of estrogen on serum NO and eNOS mRNA expression in aorta in different aged ovariectomized rat.Methods:Thirty young (3month) female wistar rats and thirty aged female wistar rats (18-20month) were randomly divided into3groups, respectively, n=10:Sham, OVX, OVE. Sham (sham operated, saline0.04ml im every three day), OVX (ovariectomized, saline0.04ml im every three day), OVE (ovariectomized+Estradiol benzoate10ug im every three day). The concentration of NO and the expression of eNOS mRNA in aorta were measured and the organ morphological changes of the aorta was observed after4month.Results:1.The serum NO level was higher in young sham group, OVX group, OVE group compared with old sham group, OVX group, OVE group (p<0.05, p<0.05, p<0.05, respectively). After ovariectomized, the serum NO level decreased in young OVX group (p<0.05), but not in old OVX group. The NO snthysis increased in OVE group compared with OVX in young and old rats with estrogen treatment (p<0.05, p<0.05, respectively).2. The expression of eNOS mRNA in aorta was higher in young sham group, OVE group compared with old sham group, OVE group (p<0.05, p<0.05, respectively), but there was no significant different between young OVX group and old OVX group (p=0.462). After ovariectomized, the expression of eNOS mRNA decreased in young OVX group(p<0.05). Estrogen treatment upregulated eNOS mRNA expression in young OVE group compared with OVX group (p<0.05). But there were no significant different in the eNOS mRNA expression in aortas from aged rats between sham group, OVX group, OVE group (p=0.579).Conclusions:1. Estrogen treatment increased serum NO level with upregulating eNOS mRNA expression in young ovariectomized rats.2. Estrogen treatment increasing serum NO level was not achieved through the increased of the eNOS mRNA expression in old ovariectomized rats.3. The serum level of NO and eNOS mRNA expression gradually decreased with ageing.
