Part1:The expression of GRP78in esophageal squamous cell carcinoma was determined using immunohistochemistry analysis.Objective:To investigate the expression of glucose regulated protein78(GRP78) in esophageal squamous cell carcinoma, atypical hyperplasia, and normal esophageal squamous epithelial tissue, and the role of GRP78in the development of esophageal squamous cell carcinoma.Methods Esophageal squamous cell carcinoma, atypical hyperplasia, and normal esophageal squamous epithelial tissues from90patients with esophageal squamous cell carcinoma were employed in the study. The expressions of GRP78in different tissues were detected using immunohistochemistry analysis. In addition, the relationships between GRP78expression and gender, age, differentiation, invasion, disease stage, and lymphoid node metabasis were analyzed.Results The positive ratio of GRP78expression in normal esophageal squamous epithelial tissue, atypical hyperplasia, and esophageal squamous cell carcinoma were7.78%,85.56%, and47.78%. The GRP78expressions in different tissues were significantly different (p<0.01). Negative GRP78expression was more often found in older patients. GRP78expression was positively correlated with differentiation, disease stage and lymphoid node metabasis.Conclusion GRP78may play an important role in the transition of normal esophageal squamous cells to malignant cells. To determine the expression of GRP78may be helpful for the prevention and early diagnosis of esophageal squamous cell carcinoma. Part2:The expression of GRP78mRNA in esophageal squamous cell carcinoma was detected using RT-PCR.Objective:To investigate the expression of GRP78mRNA in esophageal squamous cell carcinoma.Methods:The expression of GRP78mRNA in cancerous tissues and paracancerous tissues from30patients with esophageal squamous cell carcinoma were analyzed using RT-PCR analysis.Results:GRP78mRNA was positive in56.67%(17/30) cancerous tissues and26.67%(8/30) paracancerous tissues of esophageal squamous cell carcinoma. The expression of GRP78mRNA was significantly higher (p<0.05) in cancerous tissues compared with paracancerous tissues.Conclusions:GRP78may be involved in the pathogenesis of esophageal squamous cell carcinoma and may be a potential tumor maker of the disease.
