Objective: To observe the effect of resveratrol on streptozotocin-induced diabetic rats inretinal gene expression of SIRT1and the related factors.explore its possible mechanisms ofdiabetic retinopathy.Methods:The15normal rats as normal control group(group NC),57rats induced by STZsuccess only become diabetic model rats,random number table method was applied to themwere randomly divided into diabetic model(DC) group,low Res dosage of medicine(RL)group,and high Res dosage of medicine (RH)group,each group had19rats.Each group wasgiven different dose of resveratrol by gavage, continuous12weeks,concept of hematoxylin-eosin staining in the rat retina tissue pathological changes,immunohistochemical staining toobserve P53and NF-κB protein expression,use RT-PCR to assay SIRT1、P53and NF-κBmRNA expression and use ELISA method to detect SOD, GSH-PX activity in the rat retina,toobserve the effects of resveratrol on diabetic rats the above indicators.Results:Weight results showed that:compared with group NC,group DC weight loss;therats in group RL are heavier than group DC and lighter than group NC(P<0.05);The weights ofrats in group RH was no evident difference compared with group NC and group RL(P>0.05)Fasting blood-glucose results showed that: compared with group NC, the fasting blood-glucose of group DC elevated; the fasting blood-glucoses of group RL and group RH are lowerthan group DC,higher than group NC; the fasting blood-glucose of group RH is lower thangroup RL(P<0.05).Histopathological examination showed that the structures of the rats retinas of group NCare clear.The retinas of group DC become thin, structures are damaged,numbers of ganglioncells are reduced.Compared with group DC,the situations of group RL improved slightly.Thestructures of group RH are almost normal.Immunohistochemical staining results showed that:P53protein expression:thenumber of yellow or brown granules in the nucleuses of rats retinas of group DC(103.86±8.07) is more than that of group NC(9.14±2.04), the numbers of group RL(63.71±6.75)and group RH (15.57±1.99)are less than that of group DC(P<0.05). NF-κB proteinexpression:the number of yellow or brown granules in the nucleuses of rats retinas of group DC(49.70±9.50) is more than that of group NC(3.20±0.92), the numbers of groupRL(22.40±4.09) and group RH (7.50±1.58)are less than that of group DC(P<0.05).RT-PCR results showed that:The expression of SIRT1mRNA in the retina of group DC(1.23±0.03)is lower than group NC(1.50±0.10), the expressions of SIRT1mRNA inthe retina of group RL(1.27±0.03) and group RH (1.39±0.05)are higher than group DC,and lower than group NC; and the expression of P53mRNA of group DC(1.50±0.08) ishigher than group NC(0.95±0.11), the expressions of P53mRNA in the retina of group RL(1.43±0.11)and group RH (1.03±0.09)are lower than group DC,and higher than groupNC; and the expression of NF-κB mRNA of group DC(1.04±0.034) is higher than groupNC(0.22±0.01), the expressions of NF-κB mRNA in the retina of group RL(0.88±0.03)and group RH (0.74±0.01)are lower than group DC,and higher than group NC;thedifference between the expressions of SIRT1、P53、NF-κB mRNA in the retina of group RLand group RH have statistical significance(P<0.05).The activity of SOD and GSH-PX results showed that: the activity of SOD and GSH-PXin the rats retinas of group DC is lower than group NC;the activity of SOD and GSH-PX inthe rats retinas of group RL is higher than group DC;the activity of SOD in the rats retinas ofgroup RH is higher than group DC(P<0.05);the activity of GSH-PX in the rats retinas ofgroup RH has no significant difference compared with group DC(P>0.05).Conclusion: Resveratrol has certain protective effect on the damage of diabetic ratsretinal.It may be related to the mechanism of resveratrol increases SIRT1expression in theretina,affecte by the oxidative stress reaction,thus inhibit the cell apoptosis of retina andimprove the development of diabetic retinopathy...
