Objective:We try to explore the mechanism of Wnt pathway intervening airway remodeling by detecting β-catenin, RhoA and Wnt5a proteins expression of three Wnt pathways in small airway fibroblasts of rats with Chronic obstructive pulmonary disease (Chronic obstructive pulmonary diseases, COPD), and the main factor (TGF-beta1, PDGF, MMP-9)of COPD airway remodeling regulating by the Wnt pathway. We also try to use the traditional Miao medicine to treat the rats with Chronic obstructive pulmonary diseases(COPD)and detect the β-catenin, RhoA and Wnt5a proteins expression in airway fibroblasts in order to explore the mechanism of Wnt pathway regulating by the traditional Miao medicine intervening airway remodeling.The methods:1、The Wnt pathway intervening airway remodeling of rats with COPD: The100male SD rats (100±20g), aged6weeks, were randomly divided into normal group (20) and COPD model group (60). Then we established COPD model by using the method of smoking cigarettes and injecting Lipopolysaccharide into trachea of rats.2. After that, we used ELISA to detect the levels of cytokines TGF-β1, PDGF, MMP-9in supernatants of airway fibroblasts, and used tissue adherent method to culture airway fibroblasts3. β-catenin, RhoA, Wnt5a gene in normal and COPD model group were detected by Western blot to find the different expression in airway fibroblasts. In order to Studying the correlation between the expression levels of β-catenin, RhoA, Wnt5a with TGF-β1, PDGF, MMP-9.4. The Miao medicine, Myrsinaceae and Lespedeza cuneata.intervening the Wnt pathway:to detect protein expression of β-catenin, Wnt5a, RhoA in airway fibroblasts after rats with COPD being treated with the Miao medicine, Myrsinaceae and Lespedeza cuneata. Statistics:The data were analyzed by SPSS18.0. Measurement data were expressed as mean±standard deviation (x±s), the two compared groups using independent sample t test, the three compared groups using ANOVA and SNK-q test, the correlation analysis using Pearson correlation. Results:1. the levels of cytokines TGF-β1, PDGF, MMP-9in supernatants of airway fibroblasts improved.2. The expression of β-catenin, RhoA and Wnt5a of three Wnt pathways in rats with Chronic obstructive pulmonary disease increased3. The relationship between the three different ways of Wnt pathways and the levels of cytokines TGF-β1, PDGF, MMP-9in supernatants of airway fibroblasts were positive correlation.4. In Miao medicine group,β-catenin, RhoA expressing in low, while Wnt5a expressing in high.Conclusions:1. Through the study we find that the three cytokines TGF-β1, PDGF, MMP-9in supernatants of airway fibroblasts are the key factors of airway fibroblast proliferation in airway remodeling of rats with COPD. While the Wnt pathway has do to with the tissue fibrosis, and it could regulate TGF-β1, PDGF, MMP-9. Thus, we could conclude that the Wnt pathway plays an important role in airway remodeling of COPD rats.2. The study shows that the Miao medicine, Myrsinaceae and Lespedeza cuneata.could inhibit the β-catenin, RhoA of the Wnt pathway...
