[objectives] This paper presents a methodological analysis of searching for expressed differential proteins as well as discussing the material foundation of three different TCM types by comparing the protein of patients who suffering from OAK(osteoarthritis of the knee) of different TCM types with the protein in synovial fluid of traumatic synovitis of knee joint.[methods]15samples were chosen from2011January1st to2013January31st at Guiyang college of Traditional Chinese Medicine in order to compare three different TCM types of OAK patients who were separated into groups before and after treatments according to the TCM syndrome differentiation:Deficiency of kidney (5samples)、Yang deficiency and cold coagulation type (5samples)、blood stasis type (5samples)、patients of traumatic synovitis of knee joint as reference (5samples), Extraction of synovial fluid were done separately and iTRAQ analysis was made in order to find the differential protein, while patients in treatment groups would be treated with appropriate prescriptions according to the result of TCM syndrome differentiation.[results]251protein groups were found during the experiment,246protein groups had quantitative information through7channels. Comparisons were done between these7groups as Aa with D、Ba with D、 Ca with D、Aa with Ab、Ba with Bb、Ca with Cb to analyses the differential property and index.32differential proteins from all treatment group and reference group (p<0.01) were found through this comparison and9of them might be Osteoarthritis and synovitis proteins, they were listed below: histone H4、histone H2A、S100calcium binding protein A8、fibrinogen γ、fibrinogen α、cDNA (FLJ92148)、C4bconjugated protein、Part of transketolase variant、Lumican gene, the last one, Lumican gene was the "high" expressed peaks of protein while the rest were "low" expressed. The containment of HIST1H2BC and MPO in group Aa was less than group D and group Ab, the containment of Serum amyloid P protein and Apolipoprotein C-I variant in group Ba was less than group D and group Bb, the containment of Histone H2A、G reactive protein and fibrinogen a in group Ca was less than group D and group Cb.[conclusions] The differential proteins between OA(osteoarthritis) and traumatic synovitis of knee joint, synovial fluid might be9proteins listed as following:histone H4、histone H2A、S100calcium binding protein A8、fibrinogen γ、fibrinogenα、 cDNA (FLJ92148)、C4bconjugated protein、 Part of transketolase variant and Lumican gene, which can be labeled to research diseases. HIST1H2BC and MPO might be the differential protein of Kidney deficiency; Serum amyloid P protein and Apolipoprotein C-I variant might be the differential protein of Yang deficiency and cold coagulation; Histone H2A、C reactive protein and fibrinogen α might be the differential protein of blood stasis. Those three differential proteins according to the Bone inflammatory joint varied of different TCM types provided the material foundation for The classification and treatment of clinical Chinese Medicine. The synovial fluid in OA of Kidney deficiency and Yang deficiency and cold coagulation has a fairly good Protein interaction.
