According to relevant researches, tea polysaccharide possesses a variety of bioactivity, for instance, anti-tumor, antithrombosis, anti-radiation and various of physiological functions including enhance the body immunity, meanwhile, it has less sideeffect, which can be used as a functional food in pharmaceuticals industry for auxiliary treatment of chronic disease.This research selected keemun black tea originating from Qimen county, Anhui province of China. Polysaccharide obtained from keemun black tea (KBTP) by means of water boiling and ethanol sedimentation and we firstly used HPLC to analyze the monosaccharide composition of KBTP. Simultaneously, further research the biological activity in vivo and vitro about antioxidant, hepatoprotection and adjust the metabolism of glucose and fat. Results displayed that KBTP is a kind of acidic heteropolysaccharide composing of nine kind of monosaccharides, the corresponding percentage is as follows, D-Mannose (12.1%), D-ribose (10.4%), L-rhamnose (3.1%), D-glucuronic acid (2.7%), D-galacturonic acid (6.7%), D-glucose (23.5%), D-xylose (2.8%), D-galactose (19.2%) and L-arabinose (19.5%). Appraisal system of antioxidant activity showed that KBTP possesses remarkable free radical scavenging activity, which appeared a fine dose-response relationship. Whereas, in comparison with their IC50, the antioxidant activity of KBTP just below the level of positive medicine (BHT). In short, KBTP is a natural antioxidant which has observably radical scavenging activity.Research on animal (mice) mode of acute and chronic liver injury was conducted by administration of a given dose of KBTP and measure the biochemical index of its serum and tissue, the results revealed that KBTP could adjust enzymes activities and prompted correlation enzymes metabolism tend to relative normal physiological level. That is, it can improve the change of animals’weight and HI caused by liver damage and induced the serum AST and ALT levels decrease to normality as well as the tissue SOD, GSH, MDA,8-iso-PGF2a and ADH. Meanwhile, the histopathological section observation showed that KBTP hepatoprotection. In addition, the research of HFD+STZ induced diabetes type2revealed that administration of KBTP for8weeks in sequence could dramatically improve mice blood glucose and lipid levels, such as FFA, HDL-C, LDL-C, TG, TC and so on, and taken on a dose-dependent. In present study, KBTP have been revealproved to possess significant antioxidant activity and hepatoprotection for the first time as well as improvement of the abnormity metabolism of glycolipid caused by type2diabetes, which provide the theoretical basis for further development of keemun black tea in the field of natural health protection materials.
