Objective:To observe the effect of heme oxygenase-1(HO-1) towards transforming growth factor-β1(TGF-β1) and monocyte chemotactic protein-1(MCP-1) in lung tissue of rats with emphysema.Methods:Eighteen wistar rats were equally randomized to three groups:control and unexposed group(C),smoke exposure group(SM) and HO-1treated group(H). Rats emphysema model were established by exposing them to the environmental smoke daily. Observe pathologic changes of the lung tissue,measurement and classification the total and differential cell counts of bronchoalveolar lavage fluid(BALF) in all three groups. The levels of MCP-1in BALF and serum were measured by ELISA.The expressions of HO-1and TGF-(31were observed immunohisto chemically.Results:1.The pathological morphological changes:The structure of lung tissue in rats were normal in group C; All rats of SM group showed wide inflammatory changes in bronchiole and emphysema, which was accorded with clinical manifestation of the emphysema. The airway inflammation and emphysema of group H significantly reduced as compared with that of SM group.2.Measurement and classification cells:Compare with group C, the numbers of total cells and neutrophils, macrophages in BALF of group SM and group H were increased(P<0.05); compared with group SM, the numbers of total cells and neutrophils, macrophages in BALF of group H were decreased? but it was still higher than that of group C(P<0.05).3.The levels of MCP-1in BALF and serum:compared with group C, the levels of MCP-1of group SM in BALF were obviously increased(P<0.05), the levels of MCP-lof group SM in serum were increased, but the increase was not obvious (P>0.05).Compared with group SM, the levels of MCP-1of group H in BALF were decreased (P<0.05), the levels of MCP-1in serum were decreased, but the decline was not significant (P>0.05).4.Compared with group C, the expressions of HO-1、TGF-01of group SM were increased (P<0.05); Compared with group SM, the manifestations of HO-1of group H were increased (P<0.05), the expressions of TGF-β1of group H were decreased,but it was still higher than that of group C (P<0.05)5.Correlation analysis:in group SM, the levels of MCP-1in the BALF were positively correlated respectively with the number of macrophages and neutrophils (r=0.816,P<0.05r =0.873,P<0.05); by contrast, the levels of MCP-1in the serum of group SM were not positively correlated respectively with the number of macrophages and neutrophils in BALF (r=0.590, P>0.05r=0.643,P>0.05). The TGF-β1OD value of group SM were positively correlated relationship respectively with the number of macrophages and neutrophils in BALF (r=0.836, P<0.05r=0.832, P<0.05)Conclusions:There are close correlations between TGF-β1, MCP-1and emphysema. The early intervention of HO-1may inhibit lung tissue the infiltration of macrophage、neutrophil and the expressions of MCP-1and TGF-β1, which has certain protective effect to emphysema.
