Anxiety disorder is a common mental illness and has a strong influence on society. There are usually awareness in advance, but inevitable and vicious incident to trigger an expected response, such as fear, anxiety, nervous disorder as the main performance, and often accompany with heart palpitations, sweat, respiratory distress and plant nerve dysfunction. In China, anxiety has become a common disease and frequently encountered disease, which make a serious influence on life quality of resident. The traditional Chinese medicine has drawn researchers’ attentions due to its effect on the regulation of mood.Flos Albizziae (FA) is the dry flowers or flower buds of Albizzia julibrissin Durazz. It is often used for the treatment of uneasy, melancholy and insomnia. Antidepressant, hypnotic and sedative effects of FA had been reported, whereas the anxiolytic effect of FA had not been studied. The research about FA has many problems such as the ambiguity pharmacodynamic material basis. In order to lay the foundation for further study, it is necessary to clarify its anti-anxiety efficacy material base and effective mechanism.This research used pharmacological models to evaluate the anxiolytic effect of the water extract and total flavonoids of FA, and quercitrin. Purification of total flavonoids and its content determination methods were discussed in order to identify the material basis of FA. The anxiolytic effect of quercitrin had been eveluated by pharmacological models and the mechanism of its anxiolytic activity was also explored.The archieved results are as follow:1 Anxiolytic research of FAThe EPM test and LDB test were applied to eveluate the anxiolytic effects of water extract and alcohol extract of FA. In EPM test, the alcohol extract of FA group which dosage is 6g/kg significantly increased OE% and OT%. In LDB test, this group also increased the time spend in light box and entries into light box. The alcohol extraction of FA showed anti-anxiety activities on those two pharmacological models.2 Anxiolytic researches of the total flavonids of FA2.1 Purification of total flavonoids of FAThis resercach investigated how to use macroporous resin to purify the total flavonoids of FA. The single factor influence on the purification of total flavonoids were investigated, such as resin type, concentration of sample quality, pH value, volume flow, water volume, concentration of ethanol and the times of purification.2.2 Explore the content determination methods of total flavonoidsHPLC and UV spectrophotometry methods were been built to determinate the content of total flavonoids of FA, The results showed that both methods were suitable for content determination of total flavonoids.2.3 The anxiolytic effect of the total flavonoids of FAThis research used the EPM test and LDB test to eveluated the anxiolytic effects of the total flavonoids of FA. In EPM test, the alcohol extract of FA group which dosage is 2.5g/kg significant increased OE% and OT%. In LDB test, this group also increased the time spend in light box and entries into light box. The experimental results showed that the total flavonoids of FA showed anxiolytic activities on those two pharmacological models.3 The anxiolytic effect of quercitrin3.1 Pharmacodynamic studies in miceThis research used the EPM test, the LDB test, the MB test, the OFT test and spontaneous test to eveluate the anti-anxiety effects of quercitrin and its influence on mice’spontaneous activites. In EPM test, the quercitrin group which dosage is 5mg/kg significant increased OE% and OT%.In LDB test, this group also increased the time spend in light box and entries into light box. The open field test and spontaneous test show that quercitrin did not affect the spontaneous activities of mice. The experimental results showed that quercitrin showed anxiolytic activities without affect spontaneous activities on those pharmacological models.3.2 Exploration of effective mechanism3.2.1 Effects of quercitrin on 5-HT1A receptorGABAA/benzodiazepine mediation was evaluated by pretreating the mice with the GABAA receptor antagonist flumazenil before the EPM test and LDB test. Serotonergic mediation was studied by pretreating the mice with WAY 100635, a serotonin 5-HT1A receptor antagonist before the behavioral experiments. The anxiolytic effect of quercitrin was blocked by WAY 100635, whereas, those activities were not affect by flumazenil.3.2.2 Effects of quercitrin on monoamine neurotransmitterThe concentration of monoamines (5-HT and DA) and their metabolites (5-HIAA, DOPAC and HVA) in brain of mice were analyzed by the HPLC-ECD. The concentration of monoamines and their metabolites in brain declined after the treatment of quercitrin.4 ConclusionThe pharmacological models were used to evaluate the anxiolytic effect of the water extract of FA, total flavonoids of FA and quercitrin. Quercitrin was found to be the active compound in FA, the anxiolytic effects of quercitrin might be mediated by 5-HT1A receptors but not involved with benzodiazepine site of GABAA receptors. The result of neurochemical studies suggested that the anxiolytic effect of quercitrin might mediated by decrease in synthesis and release of monoamine neurotransmitters.
