Study On The Effect Of Matrine On Apoptosis Protein Of Cervical Cancer Cell Line

[Objective]Cervical cancer is one of the most common gynecological malignancy,early diagnosis and timely treatment of Cervical lesions have a great significance for the prevention of cervical cancer.In recent years,as the reports about TCM treatment of cervical lesions are more and more,the advantages of TCM are more and more obvious.Collect the literature of clinical study about the local treatment of the human papillomaviruses (HPV) infection, inflammation and the cervical intraepithelial neoplasia (CIN) with TCM published last ten years,finish used herbs and study using drugs regularity of local treatment.Take one conventional TCM as example,further experimentally study were carried out to study the effects of matrine on cervical cancer SiHa cells bax, bcl-2 protein expression and to investigate the mechanism.[Mehtods]1.Literature Study:Collect literature about the local treatment of the human papillomaviruses (HPV) infection, inflammation and the cervical intraepithelial neoplasia (CIN)with TCM from January 2004 to August 2014 in CNKI,arrange the herbs and establish excel database.The retrospective study was conducted to obtain the distribution of the medicine’s category,tastes of herbs and the channel-tropism of medicine by statistical analysis.2.Experimental Study:Based on the clinical research,Changes in cell morphology was observed under an inverted microscope after matrine acting on the cervical cancer SiHa cells.MTT assay influence of matrine on cervical cancer SiHa cell proliferation and obtain the appropriate concentration and time for matrine acting.The changing of the expression level of Bcl-2 and Bax was observed by the Western Blot.[Results]1.Literature Study:(1)According to sources of information and retrieval methods were obtained 138 documents,which met the inclusion criteria of the literature 110. A total of 112 prescriptions was collected.(2)Prescriptions were collected for analysis of frequency.There are 144 kinds of herbs were used in the local treatment of cervical lesions with TCM, and the usage frequency was 690.Borneol, Phellodendron and Sophora flavescens Ait are the top three overall.Of the top three,Sophora single herb topical treatment of cases of cervical lesions up.(3)Fourteen categories are summarized by pharmic function.Heat-clearing drugs and huoxue huayu herbs are the top two. The Heat-Clearing Damp-Drying Chinese Herbsmedicine is the most commonly used of all the Heat-clearing drugs.(4)According to the tastes and the channel-tropism of herbs,he herbs that appear most are in sequence of the bitter and cold herbs and the heart meridian herbs.2.Experimental Study:(1)After matrine acting on the cervical cancer SiHa cells,morphological changes of apoptotic cells occurred,such as cell shrinkage and rounded, cytoplasm concentrated and blistering.(2)After 24,48,72 hours of cell culture, the growth of the control group was active, and the proliferation of each matrine group were inhibited to varying degrees.The matrine more and more obviously inhibit cervical cancer SiHa cell apoptosis with the prolonged of action time and the increase of action concentrations.after matrine acting for 72 hours, the cells were almoat all dead.The inhibition rate is calcultated based on the OD of each group after atrine acting for 24 hours and 48 hours,the difference of the groups is statistically significant(P<0.05).Analyze the inhibition ratio of each group, and select 24 hours as action time and 2.0mg/ml、2.5mg/ml as action concentrations to conduct Western blot.(3)In the three groups of control group,2.0mg/ml group and 2.5mg/ml group,the expression level of Bcl-2 decrease gradually and the expression level of Bax increases gradually. the difference of the groups is statistically significant(P<0.05).[Conclusion]1.Literature Study:From the perspective of traditional Chinese medicine,the accumulation of the Dampness-heat lead the corruption of form-quality and the injury of blood vessels,then it often causes Cervical Lesion,such as the human papillomaviruses(HPV)infection,cervicitis and the cervical intraepithelial neoplasia (CIN).Dampness-heat, poison and stasis are the key of the lesions.So,cleaning heat and drying humidity are applied in the treatment,and also activating blood and eliminating stasis.Besides,local administration by facilitating the drug to reach the focal location is with good results.2.Experimental Study:The matrine could inhibit cervical cancer SiHa cell proliferation and induced cellular apoptosis.Decreasing in the expression amounts of Bcl-2 and increasing in the expression amounts of Bax to reduce the ratio of Bcl-2 to bax may be one of the mechanism.
