Objective:To investigate the effects of matrine and combined with cisplatin on expression of TSLC1gene in human cervical cancer SiHa cell and its sensitive effect on SiHa cells to cisplatin, and to search for a perfect way in the treatment of cervical carcinoma with medicine.Methods:SiHa cells cultured in vitro were treated with matrine only, cisplatin only, matrine combined cisplatin as long as48hours, and then were detected the inhibition of cells by MTT.The expression of TSLC1mRNA were detected by RT-PCR.Results:The inhibitory effect of matrine only, cisplatin only, on the growth of SiHa cell lines was in a time dependent manner. The apoptosis rate and TSLC1mRNA expression were obviously increased following the increasing concentration (P<0.01). respectively. When matrine combined with cisplatin, the inhibition rate much higher than only matrine or cisplatin, used(P<0.05).Conclusion:Matrine and cisplatin could inhibit proliferation and induce apoptosis of SiHa cells, which may be related to the upregulation of TSLC1mRNA expression as well as increase the sensitivity of Hela cells to cisplatin.
