Cortex Moutan, Rhizoma Alismatis and Yam are commonly used in clinical medicine.Chinese Pharmacopoeia (ChP) 2010 recorded the quality standards of these three herbs and their processed pieces, including Cortex Moutan, Rhizoma Alismatis, Rhizoma Alismatis processed with salt, Yam and Yam processed with wheat bran. Because of the single index detection and the single compoenet, we can not fully reflect the quality of those herbal pieces the above mentioned. In this paper, on the basis of the Chinese Pharmacopoeia 2010 and the quality standard of Beijing 2008, those herbal pieces were studied by following ways in order to improve the quality control level of the current quality standards. 1. Research on TLC identification by C18 TLCEstablishing five herbal pieces qualitative identification by using C18 TLC identification istead of Silica gel TLC for the first time, including Cortex Moutan, Rhizoma Alismatis, Rhizoma Alismatis processed with salt, Yam and Yam processed with wheat bran. Compared with other current quality standards, Cis TLC could get more information. Pretreatment methods used in this study are more convenient and fast, expanding agent is relatively safe and increasing the number of index components, improves the accuracy and safety of identification.2. Research on HPLC fingerprintHPLC method was developed to establish a fingerprint of Cortex Moutan, Rhizoma Alismatis and Rhizoma Alismatis processed with salt. There are 16 characteristic peaks in the fingerprint of Cortex Moutan. The similarity of 17 batch of Cortex Moutan was higher than 0.97. There are 9 characteristic peaks in the fingerprint of Rhizoma Alismatis samples.the similarity of 21 batch of Rhizoma Alismatis samples was higher than 0.85.There are 10 characteristic peaks in the fingerprint of Rhizoma Alismatis and 13 characteristic peaks in the fingerprint of Rhizoma Alismatis processed with salt. The similarity of 14 batch of Rhizoma Alismatis was higher than 0.83, while the similarity of 7 batch of Rhizoma Alismatis processed with salt was higher than 0.96.The HPLC-ESI-MS/MS was used to identify those main characteristic peaks in the fingerprints. There are 17 charcteristic peaks in the fingerprint of Cortex Moutan have been identified. There are 6 charcteristic peaks in the fingerprint of Rhiizoma Alismatis and Rhizoma Alismatis processed with salt have been identified. By establishing the fingerprints of Cortex Moutan, Rhizoma Alismatis and Rhizoma Alismatis processed with salt, and further identified the charcteristic peaks, we can get more informations of HPLC fingerprints.3. The simultaneously determination of multi-target ingredintsThe method for content determination of Gallic acid, Paeoniflorin,1,2,3,4,6-pentagalloy glucose and Paeonol in Cortex Moutan by HPLC was established. The method for content determination of Alisol A, Alisol A-24-aceate and Alisol B-23-aceate in Rhizoma Alismatis by HPLC was established. The methods established in this study have high precision, good stability and good repeatability. The accuracy is in the range of 95%~100%. The method used in this research is accuraty and reliability. It could used as the quality standards of Cortex Moutan, Rhizoma Alismatis and Rhizoma Alismatis processed with salt.4. Research on water decoctionThe method of content determination of the water decoction of Gallic acid, Paeoniflorin, 1,2,3,4,6-pentagalloy glucose and Paeonol in Cortex Moutan by HPLC was established. After boiling in the water, the content of gallic acid increased significantly, the content of 1,2,3,4,6-pentagalloy glucose and Paeonol decreased significantly, the content of Paeoniflorin didn’t change significantly.The method of content determination of the water decoction of Alisol A, Alisol A-24-aceate and Alisol B-23-aceate in Rhizoma Alismatis by HPLC was established. After boiling in the water, the content of Alisol A, Alisol A-24-aceate and Alisol B-23-aceate decreased significantly. This study provides some theoretical basis for the practical application of cllinical Chinese medicine.5. Research on extraction and separationBy using Silica gel column chromatography,2 fatty acids are separated from Yam. To identify the strcture, ESI-MS,’H-NMR and 13C-NMR are used in this study. The structures are 12,15 heneicosadienoic acid and stearic acid.
