Juncus setchuensis Buchen. is widely distributed in China. Its root, stem pith and whole plant are locally served as medicinal use. It has similar activities as Juncus effusus L.. Our previous study showed some phenanthrenes from Juncus effusus possess anxiolytic ffect. In order to study the structure-activity relationship of phenanthrenes. This study focus on isolation and elucidation of phenanthrene compounds from the aerial part of Juncus setchuensis.This dissertation consists of two main parts:The review part was divided into three topics as following:1. Phenanthrenes from the genus of Juncus in recent years.2. Research advance of Juncus setchuensis.3. Research Progress on techniques for chemical sudy of traditional Chinese Medicine.The experimental part was divided into two topics, including isolation and structure elucidation of chemical constituents from Juncus setchuensis and summarization of identification characteristics of phenanthrenes by TLC、HPLC、UV、IR and MS.1. According to UV and NMR sepctrosopies, phenanthrenes from the EtOAc fraction of the aerial part of Juncus setchuensis was enriched by a flash chromatography.2. Seventeen compounds were isolated from the aerial part of Juncus setchuensis Buchen. by a combination of various chromatographic techniques, and fifteen compounds were identified to be effusol、juncusol、juncuenin B、dehydroeffusol、dehydrojuncusol、 dehydrojuncuenin B、juncuenin D.2-methoxyl-7-hydroxyl-1-methyl-5-vinyl phenanthrene、 2-hydroxyl-7-carboxy-1-methyl-5-vinyl-9,10-dihydrophenanthrene、2-hydroxyl-7-carboxyl-1-methyl-5-vinyl phenanthrene、luteolin、vanillic acid、daphnetin、p-coumaric acid and ferulic acid. Daphnetin was isolated from Juncus plants for the first time. 2-Methoxyl-7-hydroxyl-1-methyl-5-vinyl phenanthrene、2-hydroxyl-7-carboxy-1-methyl-5-vinyl-9,10-dihydrophenanthrene、2-hydroxyl-7-carboxyl-1-methyl-5-vinyl phenanthrene、 luteolin、vanillic acid and ferulic acid were isolated from Juncus setchuensis Buchen. for the first time.3. Summarize the chromatographic behavior and spectral characteristics of phenanthrenes from Juncus plants. The results is useful for rapid identification of phenanthrenes.
