Objective:The aim of this study is to evaluate the protection effect of Hope I decoction on ozone inhalation-induced injury in rats and provide experimental basis for research of hope I decoction.Methods:48 male SD rats were randomly divided into 6 groups, i.e., control, model, VE (0.0135g/kg), Hope I decoction at three dosages (0.045,0.090,0.180g/kg respectively). Rats in the latter 5 groups were exposed to ozone in an exposure cabinet (1.2±0.2 parts per million (PPM) 10h/d ×45d), and were intragastrically administrated with vitamin E or Hope I decoction during exposure. Then, the levels of super oxide dismutase (SOD), lipid peroxide (LPO), catalase (CAT), and glutathione peroxidase (GSH-Px) in rat serum were detected. And pathologic changes of lung tissue were observed by HE staining and Masson staining. Pulmonary surfactant protein A (SP-A) and nicotinamide adenine dinucleotide phosphate oxidase (NOX2) content in the lung tissue were also examined by immunohistochemical method, and total protein content of lung tissue was measured by TMT quantitative proteomic techniques.Results:1, serum index:compared with model group, SOD, CAT and gsh-px activity of all hope I groups increased, and LPO levels dropped.2, pathological indicators:(1) the lung tissue observation:compared with model group, inflammatory cells exudate appeared and fluid seepage collagen fiber content reduced in hope I groups of each dose; (2) the lung tissue immunohistochemical results:in hope I fluid of each dose, SP-A content was significantly higher than the model group, while SP-A NOX2 content was lower than the model group. (3) Quantitative proteomics:the model group was detected with significant expression changes of a total of 38 kinds of protein, and after hope I liquid intervention, up regulation of 26 kinds of protein expression and down regulation of 12 types of protein expression were shown.Conclusion:After rats inhaling ozone, antioxidant enzyme activity in serum reduced and lipid peroxide increased; Lung tissue fiber also increased with changes of several proteins content in the lung tissue, which indicates that inhaling ozone can cause lung damage in rats. After intervention therapy of Hope I liquid, antioxidant enzyme activity recovered, and lipid peroxide reduced in the serum; The lung tissue fiber content also reduced, with the change of protein content to a level close to the normal group in which rats inhaled air. Therefore, Hope I fluid may reduce ozone-induced lung tissue damage through increasing the body’s ability of resistance to oxidative stress and increasing the lung protective protein. This study is of certain positive significance in prevention and treatment of lung injury caused by ozone pollution.
