Objective:To obtain the changes of spontaneously hypertensive rats (SHR) with liver Yang syndrome model, and the effects of Tianmagouteng granule in treating the liver Yang syndrome, we observed rats with the liver Yang syndrome in two aspects including blood pressure, cerebral functional imaging.The focus of this study is to obtain objective visibility basis of changes in brain function following the liver Yang syndrome by using positron emission computed tomography (PET) technology, as well as revealing the central mechanism of Tianmagouteng granule in treating the damage of brain function following the liver Yang syndrome.Methods:1 We adopt methods of filling aconite decoction with isolated raised to model rats with the liver Yang syndrome.2 The spontaneously hypertensive rats were randomly divided into two groups:model group, treatment group, n= 8. Another 10 male SD rats were selected as the blank control group.3 After the success of modeling and after treatment were measured blood pressure, to judge the effect of Tianmagouteng granule on blood pressure of SHR.4 Cerebral functional imaging experiment of model group and treat group rats were carried out in E-plus animal PET scanner with 18F-FDG as a tracer.This experiment adopt matching method. SPM software which is the gold standard in the field of brain image is adoped to do image analysis and statistical study. We obtain brain regions with different FDG metabolism that is brain activation maps through two-sample t-test of datas beween model group (before modeling) and model group (after modeling), model group (after modeling) and treatment group (after modeling), treatment group (before modeling) and treatment group (after modeling). Therefore, we obtain objective visibility basis of changes in brain function following the liver Yang syndrome, as well as revealing the central mechanism of Tianmagouteng granule in treating damage of brain function following the liver Yang syndrome.Results:1 Results of blood pressure studies:Before the intervention, control group, model group, no significant differences between the baseline body weight of the rats in the treatment groups (P> 0.05), comparable the control group, the model group,the systolic pressure,diastolic pressure is higher; compared with before intervention, the rats in the control group systolic blood pressure, diastolic blood pressure (P>0.05); Model group and treatment group, the systolic and diastolic blood pressure changes have some significant difference (P<0.05), blood pressure prognosis of two group rats was lower than before the intervention. After treatment, diastolic blood pressure had no difference in model group, the treatment group two group, the systolic blood pressure (P> 0.05).2 Results of PET studies: â‘ SPM analysis of related brain areas between model group and treatment group rats (before modeling)and blank control group rats:The related brain areas with abnormal glucose metabolism of in model group rats are very many. After 28d modeling, The brain areas with dcreased glucose metabolism included (P<0.001):The right hemisphere, on the right side of diencephalon, thalamus, hypothalamus, basal ganglia; The left hemisphere:striatum, caudate nucleus, claustrum..The brain areas with increased glucose metabolism included (P<0.001):the left hemisphere brain region:corpus callosum;â‘¡SPM analysis of related brain areas between model group rats (after modeling) and treatment group rats (after modeling):The brain areas of treatment group rats with dcreased glucose metabolism included(P<0.001):right dorsal thalamus,dorsolateral nucleus, ventral lateral nucleus, the amygdale, left thalamus, cerebellum lobule, the original crack, crack front cone, loop-shaped leaflets;The brain areas with increased glucose metabolism included(P<0.001):right medial frontal gyrus, bilateral corpus callosum, hippocampus, left hemisphere:the dentate gyrus; â‘¢SPM analysis of related brain areas between treatment group rats (before modeling) and treatment grouprats (after modeling):After 28d experiment, the brain glucose metabolism in treatment group rats (before modeling) has no statistically difference compared with treatment group rats (before modeling) within the setting threshold(P<0.001).Conclusions:1 SHR elevated blood pressure is determined by multiple genes, similar to human disease,previous studies pointed out that Tianmagouteng granule on SHR can reduce the systolic blood pressure, but compared with the normal group, the results of this study suggest that Tianmagouteng granule on SHR systolic blood pressure and diastolic blood pressurelowering effect is not obvious.2 Liver Yang syndrone hypertension model rats is related brain regions abnormal glucose metabolism,And is closely related to the function of each brain region.Liver Yang type syndrone prehypertension brain damage is cognitive function change,This and the thalamus, hypothalamus, such as cognitive adjustment is closely related to the parts consistent abnormal glucose metabolism.The hypothalamus is the emotional responses of cerebral cortex under the highest integration center,With this experiment on the liver Yang hypertension model rat hypothalamus abnormal glucose metabolism.The frontal lobe, limbic lobe, and the. close ties between the hypothalamus,Constitutes the complex system of cognition, emotion regulation,This targeting on patients with high blood pressure and always consistent brain imaging clinical study. Tianmagouteng granule can significantly affect the liver Yang syndrone hypertension model of rat frontal lobe, corpus callosum, glucose metabolism in thalamus and basal ganglia regions and adjust its function.Numerous studies of prefrontal control functions found, that front and side of the frontal area, cingulate, parietal and premotor closely constitute a cognitive activities (such as attention, working memory) associated network,Human prefrontal damage can lead to execution ability to interfere with the task. Overall, results of this study,Form of glucose metabolism to reduce brain regions have higher effect, the elevated glucose metabolism of brain regions has lower effect.Therefore, we think, Tianmagouteng granule on liver Yang syndrone hypertension related brain regions the bidirectional regulation of glucose metabolism abnormality is one of the treatment the central mechanism of the disease.
