Objective:"Acute Cholecystitis" is the name of western disease, which basically symptoms are right upper abdominal pain, tenderness, nausea and vomiting, fever, jaundice and so on. In traditional Chinese medicine (TCM), according to the description of the symptoms, it could be named "hypochondriac pain", "gallswelling", "jaundice" or "chest". In recent years, with the changes of people’s diet, the accelerated pace of life, the incidence of this disease has a rising trend, especially in elder population. Because the elder people’s physiological function and immune system are declining, more complications and chronic are seriously impacting their health.There are rich experiences for the treatment of this disease with TCM, such as herbs, acupuncture and food therapy. As they had less side effects and could relieve symptoms, the therapy of TCM is more and more popular. Acupuncture in the treatment of acute cholecystitis have been documented, and GB34(yanglingquan) is a point of gall bladder meridian. Meanwhile, GB34 is also the xiahe point of gall bladder meridian. It was effective for the treatment of acute cholecystitis. Researches of GB34 and acute cholecystitis is not uncommon, but there were some problems in clinical and experimental studies. For example, someone was lack of comparisons with other points, some indexes are too single, and some subjects were selected improperly. All the above problems were led to the unreliability of those studies. Thus, this study attempted to establish the rabbit model of acute cholecystitis, and introduced the blood routine examination and micro B type ultrasound for the check of small animal, which aimed to explore the relationship between acute cholecystitis and GB34(yagnlingquan) by observing the changes of WBC counts and gallbladder wall thickness and its pathological changes after electroacupuncture"GB34".Thirty-two healthy adult male rabbits were randomly and equally divided into normal group, model group, GB34 group and SP9 (yinlingquan) group with 8 animals in each group. Except the normal group, the other 3 groups were made acute cholecystitis model by injecting Escherichia coli in gallbladder. The second days after the end of making model, corresponding points were needling by electroacupuncture for 7 days consecutively. The outcomes included WBC count, changes of gallbladder wall and surface, and the change of gallbladder tissue pathology.Results:(1) The establishment of rabbit model of acute cholecystitis. After injucting the escherichia coli into the gallbladder of rabbit, the results showed that WBC counts in model group were significantly higer than normal group at the 1st day of postoperative (P<0.01). At the 2nd day of postoperative, compared with the normal group, the thickness of gallbladder wall were observed significantly increase (P<0.05). The above two phenomena lasted the ninth day after postoperative. Meanwhile, at the end day of the experiment, we found that the gallbladder wall was observed severe edema, fibrous dysplasia and adhesion with surrounding tissues. At the 9th day of postoperative, Papillary epithelial of gallbladder appeared edema, and the mesenchyme was soaked by inflammatory cells, and the epithelial mucosa became shorter, partly disappeared, was-full-thickness necrosis, Which were all observed by the results of HE staining.(2) Effects of WBC counts by Electroacupuncture at GB34 on acute cholecystitis rabbits. After the treatment, compared with the model group, the WBC counts of GB34 and SP9 groups were all decreased (P<0.05), but there was no significant difference between the two groups (P>0.05).(3) Effects of the thickness of gallbladder wall on acute cholecystitis rabbits. After the treatment, compared with the model group, the thickness of gallbladder wall of GB34 and SP9 groups were all decreased (P<0.05), and the thickness of GB34 group was obviously lower than that in SP9 group(P<0.05).Conclusions:(l)It was feasible to establish the acute cholecystitis rabbit model by the injection of escherichia coli. (2) Bouth needling at GB34 and SP9 could alleviate the inflammation of acute cholecystitis rabbits, and also could restore the impaired wall of gallbladder. Thus, it seems that the treatment of EA at GB34 and SP9 was beneficial for acute cholecystitis. Besides, according to the changes of the thickness of gallbladder wall, the curative effect of GB34 maybe better than SP9.
