Purpose:Define the biological role of cyclin G1in genesis and development of breastcancer, and its impact on breast cancer cells stem cells, to explore whether cyclin G1effectsmalignant behavior of breast cancer by regulating the stem cells.Material and method:Proceed molecular cloning experiments by competentescherichia coli JM109and extraction of plasmid DNA molecules GFP and cyclin G1ofplasmid DNA molecules, and transfection plasmid DNA molecules to breast cancer cell lineMCF-7, MDA-MB–231by the PEI transfection method., applicate Western blot and real-time PCR to examin cyclinG1expression in breast cancer cells on genetic and proteic leveland proceed tumor steam cells.Apply cell scratch experiments,Transwell experiments andflow cytometry experiments to explore affect of cyclin G1in invasion and metastasis of breastcancer and the malignant behavior of chemotherapy resistance, etc. Spheroid ball experimentand realtime PCR test,under cyclin G1over-expression further analysis the effects onregulatory phenotype of breast cancer cell lines. And further investigate whether cyclin G1effects invasion and metastasis of breast cancer and chemotherapy resistance by regulating thestem cells.Results:1.Suceed to transformate and proposes the GFP and cyclin G1plasmid DNA molecule.2.Cyclin G1promote malignant biological behavior of breast cancer cell such asinvasion,metastasis, and resistance to chemotherapy,etc.3.Cyclin G1promotes breast cancer cell enriched population of stem cells andself-renewal,overexpression of cyclin G1increased gene expression ofCD133,CD90,Nanog,Bmi-1,ABCG2,KLF-4,Oct4related with breast cancer stem cellsfeature,the difference was statistically significant.Conclusion:1. Cyclin G1interfered malignant biological behavior of breast cancer cell such asinvasion,metastasis, and resistance to chemotherapy,etc. 2.Cyclin G1protein molecules mediate breast cancer cell self-renewal ability and regulatedbreast cancer stem cells feature.3.High expressioncyclin G1protein molecule in breast cancer cell lines increased geneexpression related with breast cancer stem cells feature,the expression level of cyclin G1protein molecule in breast cancer cell was closely related maliganant biological behavior ofbreast cancer cell such as invasion,metastasis, and resistance to chemotherapy,etc.4.Above all,cyclin G1possibly regulate breast cancer stem cells to influence maliganantbiological behavior of breast cancer cell such as invasion,metastasis, and resistance tochemotherapy,etc.
