Purpose:The differencein mi RNAs expression between MDA-MB-231 which was cultured by spheroid and attachment was detected by mi RNA array and find mi RNA associated TNBC.Material and method:TNBC stem cells was cultured by spheroid and the differencein mi RNAs expression was detected by mi RNA array.Using the target gene prediction software to predict the differential expression of mi RNAs which may be regulated.Results:A total of 157 mi RNAs with differences in different degree between the spheroid body-forming cells and the parental cells were detected by mi RNA micro-array and SAM software in MDA-MB-231.Among them,111 mi RNAs displayed high expression(at least>2folds) in the spheroid body-forming cells,46 mi RNAs displayed low expression(at<0.05folds).The mi RNAs that were detected by mi RNA micro-array are analyzed by bioinformatics analysis.We found that five mi RNAs(hsa-mi R-297、hsa-mi R-3188、hsa-mi R-4793-3p、hsa-mi R-34a-5p、hsa-mi R-6768-5p)show the characteristics of statistically differential expression.And bioinformatics analysis revealed that these mi RNAs are involved in tumor cell growth, proliferation, apoptosis and other biological functions. There were more than ten to hundreds of predicted target genes for each mi RNA and several genes were associated with some signaling pathways of the ‘stem cell genes’, such as Wnt/b-catenin,MAPK, NOTCH etc.Conclusion:1.We identified a profile of mi RNAs expressed differentially in MDA-MB-231,which provided a starting point to explore the functions of these mi RNAs.
