Purpose: Establishing model of ischemia anoxic brain damage in rats, withthe method of the right common carotid artery ligation combined with oxygendeprivation. Applying the method of combining acupuncture“shu”and“he”pointto intervene the ischemia anoxic brain damage in rats,with S100B、NOGO-A asexpression index to rat tissues. Using enzyme-linked immunosorbent method andimmunohistochemistry to research the expression of S100B,NOGO-A in the braintissue,in order to find out the mechanism of the improvement in neurobehavioralability of hypoxic ischemic brain damage rats with the intervention of “shu”and “he”point acupuncture method,thus to provide certain theoretical basisand experimental basis for clinical treatment of ischemic hypoxic brain injuryin children.Experimental materials and methods:1.Materials:(1)Experimental animals:Eight SPF female SD rat,pregnancy weightto300±30g (purchased from Liaoning immortality biological technologyco.LTD,animal number:qualified SCXK2010-0001)(2)Experimental drug:4%paraformaldehyde,anhydrous calcium chloride,sodiumlime,etc.2.Methods:(1)group and establishment of the model:8pregnant rats deliveredbaby rats79,79baby rats were randomly divided into intervention group(n=26)and the non-intervention group(n=27) and made up normal group(n=26),accordingto random number table.All the intervention group and the non-interventiongroup Ischemia hypoxia group were performed right common carotid artery ligation,and then were put into an airtight container filled with8%oxygen and92%nitrogen gas mixture for2hours.3days after the surgery, The intervention grouptake acupuncture intervention on li11point on both sides,after three holes,three mile,fall the valley point,once per day,for14days.All threegroups of young rats were performed neurobehavioral development evaluationeveryday. Meanwhile,examine the brain tissue with S100B as expression index ofall three groups on the4th,7th,21st day accordingly after their born; andthose with Nogo-A on the7th,14th,21st day accordingly after their born.Results:1.In this experiment, hypoxia and ischemia group with8pregnant rats, deliveryrats79only, in the course of the experiment group and3rats died,8died innon-intervention group, the normal group without a death.2.Detection of neurobehavioral ability: righting reflex, days7later, controlgroup took significantly less time than intervention group andnon-intervention group, well results of the intervention group andnon-intervention group are very close;14days later,the intervention group tookless time than non-intervention group;hanging test,openfield test, controlgroup took obviously longer time than the intervention group andnon-intervention group.3.By comparing the intervention group,non-intervention group with the normalgroup, the quantity of S100B and Nogo-A are larger than the normal group, andthe difference has statistical significance(p<0.01); Regard to the expressionof S100B on the4th,7th day, the difference between intervention group andnon-intervention group shows no apparently statistical significance(P>0.05),well on the21st day, the results of intervention group and non-interventionare statistically different(p<0.01); regard to Nogo-A, the quantity ofnon-intervention are much larger than intervention group, and the differenceshows statistical significance(p<0.01).Conclusion:1.With the method of the right common carotid artery ligation combined withoxygen deprivation. ischemia anoxic brain injury model can be successfullyreplicated in rats. 2.The “shu”and“he” point can improve the ischemic hypoxic brain injury ratneurobehavior ability.3.The acupuncture method of “shu” and “he”point can reduce the quantity ofS100B,NOGO-A in the brain tissue of ischemia hypoxic brain injury in rats, whichcan relieve the degree of the cerebral damage and repair of brain injury.
