Objective: Through the observation the influence of electrc acupuncture Heart Sutra yuan-primary point "Shen-men" 、 Small intestine meridian luo-connecting point "zhi-zheng" and "Xin-shu" point on myocardial ischemia model of rat cerebral cortex nerve growth factor(NGF) and its receptor tyrosine kinase A(Trk A) on the expression of,Compare the differences of different acupoints acupuncture anti myocardial ischemia of braininjury,And to explore effect mechanism of acupuncture on myocardial ischemia cerebral protection.In order to to provide the experimental basis of Heart Governing Mind Theory and the difference of meridian effect,For the clinical diagnosis and cardiac insufficiency caused by brain damage to explore new treatment approach,And provide reference for clinical choose acupuncture point.Method:Select 80 healthy male SD rats,Weight220±20g,Points cage feeding,Randomly selected 10 of them were as control group,The remaining 70 only under ether anesthesia in left anterior descending coronary artery ligation method of acute myocardial ischemia model in rats,And use the Powerlab Many physiological recorder to record the electrocardiogram changes,Finally the model to replicate the success of rats were randomly divided into model group、electrc acupuncture "shen-men" group、electrc acupuncture "zhi-zheng" group and electrc acupuncture "xin-shu" group,Each group of 15 only.Every group electrc acupuncture treatment of the stimulating current of 1 m A、the frequency of 2 Hz and 15 min each stimulus.After electrc acupuncture,Death in rats for breakage,craniotomy removed intact brain tissue,isolated cerebral cortex part on the ice.For each group respectively in the rat brain cortex with immunohistochemical method and fluorescence quantitative PCR technique to detect model NGF and its receptor Trk A genes expressed in the cortex.Using Spass17.0 for statistical analysis,comparison the difference between groups of rat brain cortex model NGF and its receptor Trk A expression.Results:(1)the Powerlab electrocardiogramCompared with the control group,The rat heart rate significantly decreased after modeling,a significant elevation of ST segment,T wave inversion,Display myocardial ischemia model was duplicated successfully.(2)The results of immunohistochemical detection the electrc acupuncture in "shen-men" 、 "zhi-zheng" and "xin-shu" point of the rat brain cortex model NGF and its receptor Trk A expression①compared with control group:No statistically significant differences in the expression of model group rats cerebral cortex of NGF and its receptor Trk A(P >0.05),but the number of positive cells expression were higher than that of control group.②Compared with the model group:Number of positive expression cells each acupuncture treatment group compared with model group significantly increased,the difference was statistically significant(P <0.05).③Comparison between treatment way:Compared with the "zhi-zheng" group, "sheng-men" group express numbers are increased,no statistically significant difference(P >0.05),;the "xin-shu" group and the "zhi-zheng" group comparison is increase in the number of expression,Significant difference(P <0.01);the "xin-shu" group and the "shen-men" group comparison is increased in expression,There are significant differences(P <0.05).(3) fluorescence quantitative PCR detection of electrc acupuncture in "shen-men" 、 "zhi-zheng" and "xin-shu" point of the rat brain cortex model NGFm RNA and its receptor Trk Am RNA genes expression①Sample melting curve is a single peak,no nonspecific fluorescence,sample amplification curve of the whole parallel veracity,Inflection point clear,index period significantly、the steepness of the big,baseline flat without rising phenomenon,Show that response template no pollution,accurate quantitative,The result is credible.②Each relative expression comparison coincided with the results of immunity group detection. Compared with control group:No statistically significant differences in the expression of model group rats cerebral cortex of NGF and its receptor Trk A(P >0.05),but the number of positive cells expression were higher than that of control group.Compared with the model group:Number of positive expression cells each acupuncture treatment group compared with model group significantly increased,the difference was statistically significant( P <0.05).Comparison between treatment way:Compared with the "zhi-zheng" group, "sheng-men" group express numbers are increased,no statistically significant difference(P >0.05);the "xin-shu" group and the "zhi-zheng" group comparison is increase in the number of expression,Significant difference(P <0.01)the "xin-shu" group and the "shen-men" group comparison is increased in expression,There are significant differences(P <0.05).Conclusion:(1) Electrc acupuncture at different acupoints can improve the nerve growth factor in cerebral cortex of acute myocardial ischemia in rats(NGF) and its receptor tyrosine kinase A(Trk A)levels;And has relative specificity regulating effect on it, electrc acupuncture in "xin-shu" point better than Heart Sutra yuan-primary point "Shenmen" and Small intestine meridian luo-connecting point "zhi-zheng",but the "shen-men" point compared with the "zhi-zheng" point has no obvious difference.(2) By improving the brain cells of nerve growth factor(NGF plays) and its receptor tyrosine kinase A(Trk A) content,May be the acupuncture in the treatment of myocardial ischemia caused by one of the mechanisms of brain damage.
