Purpose:This experiment observes the effect of electric acupuncture oftsusanli and sanyinjiao acupuncture points on serum iron content, ferritincontent in duodenum, expression of liver Hepicidin mRNA, and the expression ofmRNA of DMT1and DFN1of the duodenum in rats with spleen-QI deficiency to discusswhether iron metabolism disorder is one of the pathological natures of thespleen-QI deficiency syndrome, and tries to discusses the effects and actionmechanism of acupuncture treatment for spleen-QI deficiency.Material and method: A total of40SD rats are randomly divided into4four groups of normal group, model group, acupoint group and non-acupoint groupwith10rats in each group by adopting a randomization method. After the modelingis succeed, the normal group is fed normally and continuously; modelling iscontinuously conducted on rats in model group, acupoint group and non-acupointgroup in the morning; acupuncture treatment is conducted on the non-acupointgroup in the afternoon; the model group is bound for fixing. The model groupis bound for fixing for20minutes and not subjected to acupuncture everyday;the rates in the acupoint group are bound, then tsusanli and sanyinjiaoacupuncture points are subjected to acupuncture by acupuncture needles for3-5mm, electric acupuncture apparatus is connected to conduct dilatational wavestimulation for20minutes, the intension depends on vibration of the needlehandles, and the operation is conducted once a day; the rats in the non-acupointgroup are bound, then customized non-acupoint parts are subjected to acupunctureby acupuncture needles for3-5mm, the electric acupuncture apparatus isconnected to conduct dilatational wave stimulation for20minutes, the intensiondepends on vibration of the needle handles, and the operation is conducted oncea day. After the treatment is completed, tissues of serum, liver, duodenum and thelike of the rates in each group are taken. The serum iron content in serum, andduodenum ferritin content of the rats are detected by using the ELLSA method;the expression of liver Hepicidin mRNA and the expression of mRNA of the DMT1and FPN1of the duodenum are detected by using the fluorogenic quantitative PCRmethod.Results:1. The effect of electric acupuncture of tsusanli and sanyinjiao acupuncturepoints on serum iron content of the rats with spleen-QI deficiency.Compared with the model group, the serum iron content of the rats in thenormal group is obviously reduced (P<0.01); compared with the model group, theserum iron content of the acupoint group is obviously reduced (P<0.01); thereis no statistical significance by comparing the acupoint group with the normalgroup (P>0.05); there is no statistical significance by comparing the model groupwith the non-acupoint group (P>0.05).2. The effect of electric acupuncture of tsusanli and sanyinjiao acupuncturepoints on ferritin content of rats with spleen-QI deficiency.Compared with the model group, the ferritin content of the rats in the normalgroup is obviously rised (P<0.01); compared with the model group, the ferritincontent of the acupoint group is obviously rised (P<0.01); there is nostatistical significance by comparing the acupoint group with the normal group(P>0.05); there is no statistical significance by comparing the model group withthe non-acupoint group (P>0.05).3. The effect of electric acupuncture of tsusanli and sanyinjiao acupuncturepoints on the expression of liver Hepicidin mRNA of the rats with spleen-QIdeficiency.Compared with the model group, the expression of the Hepicidin mRNA contentof the rats in the normal group is obviously rised (P<0.01); compared with themodel group, the expression of liver Hepicidin mRNA content of the acupoint groupis obviously rised (P<0.01); there is no statistical significance by comparing the acupoint group with the normal group (P>0.05); there is no statisticalsignificance by comparing the model group with the non-acupoint group (P>0.05).4. The effect of electric acupuncture of tsusanli and sanyinjiao acupuncturepoints on the expression of the DMT1gene of duodenum of the rats with spleen-QIdeficiency.Compared with the model group, the expression of the DMT1mRNA content ofthe rats in the normal group is obviously rised (P<0.01); compared with the modelgroup, the expression of DMT1mRNA content of the acupoint group is obviouslyrised (P<0.01); there is no statistical significance by comparing the acupointgroup with the normal group (P>0.05); there is no statistical significance bycomparing the model group with the non-acupoint group (P>0.05).5. The effect of electric acupuncture of tsusanli and sanyinjiao acupuncturepoints on the expression of the FPN1gene of duodenum of the rats with spleen-QIdeficiency.Compared with the model group, the expression of the FPN1mRNA content ofthe rats in the normal group is obviously reduced (P<0.01); compared with themodel group, the expression of the FPN1mRNA content of the acupoint group isobviously reduced (P<0.01); there is no statistical significance by comparingthe acupoint group with the normal group (P>0.05); there is no statisticalsignificance by comparing the model group with the non-acupoint group (P>0.05).Conclusion:1. The rats with spleen Qi deficiency syndrome have iron metabolism disorder.2. Iron metabolism disorder may be one of the pathology natures of spleen Qideficiency3. Electric acupuncture of tsusanli and sanyinjiao acupuncture points has theeffects of adjusting iron metabolism disorder of rats with the spleen Qideficiency.4. The effects of replenishing qi to invigorate the spleen of tsusanli andsanyinjiao may be completed by adjusting the iron metabolism disorder.
