Objective:To investigate the protective mechanism of Huoxuerongluo pill on Alzheimer’s disease.Methods:APP/PS1transgenic rats were randomly divided into model group, model+huoxuerongluo pill group (low-dose group, middle-dose group and high dose group), model+Donepezil group. There were5rats in each group,meanwhile KM rats were divided into normal group.After given the corresponding drugs for two months, observed the behavior of the mice in morris water maze to assess their learning and memory skills;through HE stain to examine the neuronal morphous in the hippocampus, used the westen blot and image analysis method to determine the content of Aβ、LRP-1and RAGE of the brain tissues.Result:①Morris water maze:compared with normal group and drug groups, the model group had a longer incubation and seeking period, whereas the times of across the platform is the least, there was a statistically significant difference (P<0.05or P<0.01)between them; among the APP/PS1transgenic rats,the high dose group do the best.②HE stain:the neurons in the hippocampus of the model group decreased,arranged irregularly, and the impairments of the structure were especially obvious;while the neurons of other groups were adequate, spaced regularly and got less impairment.③Westen blot:in the brain tissues,compared with normal group and drug groups, the model group had the highest content in Aβ and LRP-1, whereas the lowest content in RAGE, there was a statistically significant difference (P<0.05or P<0.01)between them; among the APP/PS1transgenic rats,the high dose group had the lowest content in Aβ and LRP-1, whereas the highest content in RAGE.Conclusion:Huoxuerongluo pill may increase the expression of LRP-1, and decrease the expression of RAGE to reduce the deposit of AP, eventually improve the learning and memory ability of APP/PS1transgenic rats, at the same time it could reduce the impairments of of the neurons in the hippocampus.
