objective:This experiment with Functional Dyspepsia (Functional Dyspepsia, FD) rats as experimental object, the observed shu stomach soup decoction on Functional Dyspepsia (Functional Dyspepsia, FD) change the type of rat intestinal neural network signaling pathways-the ICC-smooth muscle ultrastructural morphology change and the distribution of the small intestine gap junction protein Cx43protein, explore its action mechanism for the treatment of Functional Dyspepsia.Method:(1)72rats were randomly divided into control group, model group, the shu stomach soup low dose group (shu low group), shu stomach soup dose group (group) in the shu shu stomach soup, high-dose group (grace), moser will benefit group, a total of six groups, each group, of12.(2) with chronic bondage stress strain diet dishonor compound cause cause FD liver spleen deficiency rats model, the control will not be made mould.(3) building observed in rats during general behavior change and weight change.(4) continuous dosing after14d, detection of gastric emptying and small intestinal propulsion rates, take small intestine tissue do immunohistochemical double staining and fluorescence observation of Cx43protein and the ICC and the morphology of nerve fibers were observed under electron microscope in the rat intestinal tissue ENS-the ICC network ultrastructural changes of SMC.Results:(1) Compared with the control group, the rats of model group increased amount of body weight, behavior action were significantly decreased (P<0.01)(2) and the control group than that in the stomach of the rats in model group residual rate increased significantly, duodenal propulsion rate was significantly decreased (P<0.01)(3)intestinal gap connection of Cx43protein expression was significantly decreased (P<0.01) and ICC ultrastructure injury obviously, cholinergic nerve ICC-SMC network structure disorder, ICC and nerve fiber number reduced (P<0.01), fluorescence intensity was significantly reduced (P<0.01), around with axons cavitation. Synapse like junction disappeared, and Zhou Wei smooth muscle cells loosely connected, ENS-ICC-SMC network like ultra structure was destroyed.(4) compared with the model group, drug group (Gao Shu, Shu to group, mosapride group) gastric residual rate decreased significantly and duodenal propulsion rate increased significantly (P<0.01); intestinal slot connection Cx43protein expression was significantly increased (P<0.01), the normal morphology of the ICC, ICC and nerve fiber number increased significantly (P<0.01), the fluorescence intensity increased apparently(P<0.01). ENS-ICC-SMC network like ultrastructure preservation is complete.Conclusion:shuWei decoction can improve the morphology of ICC cells, to restore normal physiological function of the ICC, promote the recovery of nerve fibers in the normal morphology and function of and increase gap junctional Cx43protein expression, enhanced between ICC and smooth muscle gap junction and super micro structure complete repair of the nerves of the small intestine-ICC-SMC network, to improve gastrointestinal motility, and effective treatment of FD.
