Effects Of Electroacupuncture And Jujixu On α7nAchR And NF-κB In DU Rats

Objective:To explore whether the lower He-sea point has relative specificity when treating its Corresponding viscera disease and to observe the expression level of duodenal tissue IL-lβ, a7nAchR and NF-kB in duodenal ulcer model rats by using electroacupuncture at Xiajuxu, Zusanli, Shangjuxu, Yanglingquan and Chengjin acupoints.Methods:70SD rats (50%male and50%female) were randomly divided into7groups:Blank group, Model group, Xiajuxu group, Zusanli group, Shangjuxu group, Yanglingquan group and Chengjin group. Set up the Duodenal Ulcer rat models through injection of cysteamine hydrochloride. Then the five treatment groups respectively recived electroacupuncture with its corresponding points for10days. On11th day, duodenal tissues were collected to make DU score by eye first, and then detected the a7nAchR mRNA and NF-kB mRNA expression by PCR primers. ELISA was respectively used to check models’duodenal tissue IL-lβ level.Results:1. Morphological changes of rats’duodenal tissue in each groups:compared with normal group, all groups of duodenal tissue score were improved with statistically significant difference(P<0.01). After acupuncture treatment, when compared with model group, the ulcer score of Xiajuxu group, Zusanli group, Shangjuxu group were decreased with statistically significant difference(P<0.01or P<0.05), while the ulcer score of Yanglingquan group and Chengjin group only had downward trend but no statistically significant(P>0.05). The ulcer score of Yanglingquan and Chengjin groups were increased when compared with Xiajuxu group with statistically significant difference(P<0.05). The ulcer score of Zusanli group, Xiajuxu group and Shangjuxu group showed a trend of increase in turn but no statistically significant (P>0.05).2. The expression of duodenal tissue IL-1βamong each groups:compared with normal group, the rest six groups of tissue IL-1βlevel were improved with statistically significant difference(P<0.01). After acupuncture treatment, when compared with model group, the tissue IL-1βlevel of five acupoints groups were all decreased with statistically significant difference(P<0.01or P<0.05). When compared with Xiajuxu group, the tissue IL-1βlevel of Shangjuxu group, Yanglingquan group and Chengjin group were all improved with statistically significant difference(P<0.01or P<0.05).While the Zusanli group only had an increase trend but no statistically significant(P>0.05). 3. The expression of a7nAchR mRNA and NF-kB mRNA among each groups:compared with normal group, the a7nAchR mRNA level of model group was decreased while its NF-kB mRNA level was increased with statistically significant difference(P<0.01). After acupuncture treatment, when compared with model group, the rest five groups of NF-kB mRNA level were all decreased, while the a7nAchR mRNA level of Xiajuxu group, Zusanli group and Shangjuxu group were all improved with statistically significant difference(P<0.01or P<0.05), Yanglingquan and Chengjin group only had an increase trend but no statistically significant(P>0.05). Compared with Xiajuxu group, the NF-kB mRNA level of Chengjin group was decreased, and the a7nAchR mRNA level of Yanglingquan group and Chengjin group were all improved with statistically significant difference (P<0.01or P<0.05), while the a7nAchR mRNA level of Shangjuxu group and Yanglingquan group only had the trend of decrease, and the NF-kB mRNA level of Xiajuxu group, Zusanli group and Shangjuxu group showed an increase trend in turn but no statistically significant (P>0.05).Conclusions:1. Electricacupuncture at the lower He-sea point associated with digestive system can make treatment effect on DU model rats, and its mechanism might alleviate inflammation, improve duodenal mucosal injury by activating the cholinergic anti-inflammatory pathway.2. In contrast, electricacupuncture DU model rats at Xiajuxu point and Zusanli point, whose intervention of the inflammatory response were better than other points, which indicated that differences exist in six-fu lower He-sea points’therapy effect of DU.3. The experimental results showed that a certain relative specificity existed in the lower He-sea point Xiajuxu point when compared with the rest lower He-sea point on treating DU model rats. However, Zusanli point and Xiajuxu point had the similar effect, which might associated with some consistency existed in their intervention targets, its specific connotation and mechanism still needs further research.
