Objective:Electroacupuncture at XiaoHai and XiaJuXu acupoint on Duodenal Ulcer rat models, and then observe the effect of DU score by eyes, tissue changes under opticalmicroscope, serum TNF-a, ChAT levels and a7nAchR expression, to analyze their mechanism to which they related, briefly explaining the relative specificity of ’He points’ in ’curing viscera diseases by He points’.Method:Forty SD rats were randomly divided into4groups-control groups:blank group, model group, XiaoHai group, XiaJuXu group,10rats in each group, half male and half female. Setting up the Duodenal Ulcer rat models through injection of Cysteamine hydrochloride at the right buttock. After molding, following electroacupuncture at rat’s XiaoHai and XiaJuXu acupoint, Electroacupuncture parameters density wave (10/50Hz), a negative electrode on the right and the positive one left, the intensity between1~3mA, with limb tremor in degrees needle for30minutes once a day for10days. After treatment, make DU score by eyes first then observe the duodenal tissue changes under opticalmicroscope. ELISA and colorimetric method are used to check models’serum TNF-a and ChAT levels. Western blot is used to test a7nAchR level in Duodenal tissue. Results:1. Naked eye observe the duodenal ulcer score and observe duodenal tissue structure changes under the light microscope, it shows:①The DU score of Model group is significantly higher than the Blank group(P<0.01). The scores of Xiaohai and Xiajuxu groups are significantly lower than the Model group (P<0.01). The score of Xiajuxu group is obvious lower than Xiaohai group (P<0.01).②Under the light microscope, Pathological changes of the Blank group was not obvious;Model group rat duodenal tissue injury obviously. Obvious necrosis and larger damage, shedding were observed. There are improved changes in Xiaohai and Xiajuxu groups and the Xiajuxu group is better than Xiaohai.2.ELISA method was used to text the the serum TNF-a levels, and it shows:After modeling, each groups shows high levels of TNF-a than the Blank group, and Compare to the Model group, the levels of Xiaohai and Xiajuxu groups are significantly lower(P<0.01). and the Xiajuxu group is also lower than Xiaohai group (P<0.01).3. Colorimetric and Western blots methods are used to test the serum level of ChAT and the duodenal tissue level of a7nAchR, it shows:①Compared to the Model group, the level of ChAT for Xiaohai group shows a higher trend but with no statistical significance(P>0.05). But the level of ChAT for Xiajuxu group is significantly higher than the Model group(P<0.01) and the Xiaohai group (P<0.05).②Compared to the Model group, The levels of a7nAchR for Xiaohai and Xiajuxu groups are significantly higher(P<0.01).③The ChAT is positively correlated with the α7nAchR in every group(r=0.444, p=0.007). Conclusion:1. Comparing the results of the DU score by eyes and the duodenal tissue changes under opticalmicroscope, we find that electroacupuncture at XiaoHai and XiaJuXu acupoint have obvious treatment effects on DU rats. And Xiajuxu is much better than Xiaohai acupoint.2. Electroacupuncture at XiaoHai and XiaJuXu acupoint can obviously reduce the serum levels of TNF-a for DU rats. Which suggest that by reducing the inflammatory cytokines in serum may be the mechanism of therapeutic effect for the electroacupuncture.3.Through the analysis of the levels of TNF-α、ChAT and the the α7,nAchR, we can arrive at the conclusion that the regulation effects of inflammatory responsemay, which made by electroacupuncture at XiaoHai and XiaJuXu acupoint may be influenced by the CAP.4.The research’s results partly confirmed:the word’He points’in’curing viscera diseases by He points’referring to the lower he acupoint.
